Esta monografia investiga, descreve e analisa a adoção consentida no Brasil, mais especificadamente no Núcleo de Adoção e Estudos da Família, núcleo integrante da 2ª Vara da Infância e Juventude da Capital, na cidade do Recife em Pernambuco. Priorizamos o enfoque de uma parte integrante em processos judiciais desta modalidade, qual seja, os genitores que entregam seus filhos em adoção. Esta investigação busca compreender os motivos que impulsionam os genitores a tomar tal decisão. Nossa monografia situa o histórico da política da criança e do adolescente e a adoção no Brasil, colocando-o como análise indispensável, tendo em vista que tal retrospectiva traz à luz a discussão do novo paradigma introduzido pelo Estatuto da criança e do adolescente. O mesmo traz como princípio fundamental a Doutrina da proteção integral para todas as crianças e adolescentes considerado (a)s seres em processo de formação com peculiaridades, não podendo ser vistos como objetos de tutela, se contrapondo dessa forma a legislação antes vigente.
O Estatuto da Criança e do Adolescente visa atender a criança e o adolescente alvo nos processos de adoção, já a compreensão da problemática dos genitores, passa pelo entendimento do contexto social, econômico e cultural, que permeiam a vida desses sujeitos, considerando que tais fatores certamente determinam a decisão de entregar seus filhos a terceiros, no lugar de mantê-los consigo. Os sujeitos alvo desta pesquisa em sua totalidade pertencem às camadas pobres de nossa sociedade desprovidas por vezes dos serviços sociais essenciais. Esse contexto de precariedade das condições de existência interfere em suas histórias de vida, altera desta maneira suas decisões e possibilidades de assumir a filiação constituída.
Neste sentido, apontamos a relevância e pertinência da análise aqui apresentada contribuindo para que se conheça melhor os genitores envolvidos no processo de adoção consentida, confrontados com níveis de remuneração mínimos, precárias condições de vida e de trabalho, somados às conseqüências de fragilização emocional conseqüência da desvinculação de seus filhos. A monografia tenta tornar visível o registro da adoção consentida e retirá-la da sombra do preconceito, proporcionando um maior entendimento dos motivos que promovem a entrega de crianças em adoção no Brasil.
This monograph investigates, describes and analyzes consensual adoption in Brazil, more specifically the Adoption Nucleus and Family Studies, an integral part of the Capital of Pernambuco´s 2nd Judicial Court for Children and Adolescents in Recife, Pernambuco. Priority is given, in our approach, as an integral part of the judicial trials of this type, to those parents who give up their children for adoption. This inquiry seeks to understand the motives that stimulate parents to come to such a decision. Our monograph puts into perspective the history of this policy regarding children and adolescents and adoption in Brazil, as an indispensable analysis, and has in mind that such a retrospective brings to light the discussion of a new paradigm introduced by the Statute of Children and Adolescents. This statute brings, as a fundamental doctrine for the total protection of all children and adolescents, consideration of the process of formation of such children, each with intrinsic peculiarities, so as not to be seen as objects of guardianship, and not to go against legislation previously in force. The Children and Adolescent Statute looks to assist children and adolescents in the process of adoption, to the understand parents´ problems in a cultural, economic, and social context, and the life of these people, considering those factors which brought them to decide to hand over their child(ren) to third parties, instead of keeping them. The subjects of this research belong wholly to the poor segments of our society, deprived for times of essential social services. This context of precarious conditions of existence interferes in their life history and, in this manner, their decisions and the possibilities of assuming responsibility for their family. In this context, we look to the relevance and pertinence of the analysis here presented as contributing to know better the parents involved in the process of consensual adoption, confronted with levels of precariousness, minimal earnings and conditions of life and work, added to the consequences of emotional fragility as a consequence of their being disconnected from their children. This monograph will attempt to make visible a record of consensual adoption and remove it from shades of prejudice, providing a larger understanding of the motives which promote giving up children for adoption in Brazil.

Source text - Portuguese POLIMORFISMO: DESAFIOS E ESTRATÉGIAS EM PESQUISA, DESENVOLVIMENTO E INOVAÇÃO NA INDÚSTRIA FARMACÊUTICA
O polimorfismo de insumos farmacêuticos ativos (IFAs) e excipientes compreende um grande desafio para Pesquisa, Desenvolvimento e Inovação (P, D&I) na indústria farmacêutica, devido as propriedades de estado sólido estarem diretamente relacionadas a estrutura cristalina das moléculas. Assim, o emprego de diferentes formas polimórficas de um mesmo composto pode gerar impacto direto na formulação e produção dos medicamentos, sendo as propriedades mecânicas, a biodisponilidade e a estabilidade os pontos mais críticos a serem avaliados no desenvolvimento do produto. Neste contexto, este trabalho tem como objetivo discutir com base numa revisão da literatura os principais aspectos afetados pelo polimorfismo, bem como as estratégias e métodos empregados para caracterização, obtenção e seleção das fases polimórficas mais adequadas para a produção de medicamentos. Ademais, também serão abordados os fatores decisivos para escolha dos processos e excipientes mais apropriados para controle do polimorfismo durante a produção e armazenamento desses produtos farmacêuticos.
INTRODUÇÃO
A habilidade de um sólido existir em mais de uma estrutura cristalina a partir da mesma estrutura molecular, fenômeno conhecido como polimorfismo, compreende uma área de grande interesse e preocupação para a indústria farmacêutica (1). Isto porque diversos insumos farmacêuticos ativos (IFAs) apresentam diferentes formas polimórficas (2), as quais apresentam propriedades físico-químicas, termodinâmicas, cinéticas, interfaciais e mecânicas bastante distintas (3-5). Algumas destas propriedades, tais como a solubilidade e a estabilidade, são essenciais para o desenvolvimento bem sucedido de candidatos a medicamentos (6) e, portanto, tais divergências podem gerar impacto direto tanto no desempenho e estabilidade do produto farmacêutico, como também na capacidade de processamento do IFA e na formulação e produção do medicamento (3;7).
A cristalização dos IFAs, particularmente aqueles que possuem múltiplas formas polimórficas, figura dentre os processos mais críticos e menos compreendidos da fabricação farmacêutica. Uma série de falhas nos processos e produtos pode ser atribuída a uma compreensão e controle insuficientes dos processos de cristalização (8). Deste modo, a aparição ou o desaparecimento inesperado de uma forma polimórfica, por exemplo, pode levar a sérias conseqüências, que podem resultar em atraso do desenvolvimento do medicamento e/ou interrupção de sua produção comercial (3). Um episódio culminante, ocorrido em 1998, foi o afastamento temporário do mercado do medicamento Norvir®, devido à aparição de um polimorfo previamente desconhecido do ritonavir (6).
Como resultado, procedimentos sistemáticos de caracterização polimórfica devem ser realizados ainda nas etapas de pré-formulação (9). Portanto, uma compreensão da relação entre as propriedades de estado sólido e estruturas cristalinas das possíveis fases polimórficas de um IFA (10) possibilita a escolha da forma mais adequada para desenvolvimento de um medicamento (9) e também pode ser utilizada para aperfeiçoar as estratégias operacionais e de formulação, bem como aprimorar a concepção adequada de protocolos de estabilidade (10) a fim de evitar problemas durante ou após o lançamento dos produtos (11).
Ademais, o controle do polimorfismo tem se tornado uma das matérias essenciais no que se refere às pesquisas científicas ligadas a indústria farmacêutica (7) devido à maior parte dos produtos farmacêuticos serem formulados com excipientes e fármacos no estado sólido e forma cristalina (5; 7; 12). O desenvolvimento desses cristais moleculares em formas farmacêuticas sólidas é preferido, por razões de produção, estabilidade (5), praticidade de administração e dosagem para o paciente (4), embora também possua seus desafios, como o próprio polimorfismo (5).
Desta forma, este trabalho tem como objetivo discutir com base numa revisão da literatura os desafios advindos do polimorfismo na pesquisa, desenvolvimento e inovação de medicamentos e as estratégias e métodos empregados para caracterização, obtenção e seleção das fases polimórficas mais adequadas, bem como a escolha dos processos e excipientes mais apropriados para controle do polimorfismo durante a produção e armazenamento de medicamentos.
Tipos de Polimorfismo
As substâncias no estado sólido podem apresentar-se na forma amorfa com arranjos desordenados das moléculas e sem uma rede cristalina definida (13) ou cristalina, com moléculas dispostas segundo um padrão de repetição ordenado ao longo de toda a partícula (14). Estas substâncias cristalinas podem ser formadas por moléculas flexíveis que adotam mais de uma conformação no estado sólido (polimorfismo conformacional) ou moléculas rígidas que adotam diferentes formas de empacotamento (polimorfismo de empacotamento) (2; 5;15).
A depender do entrelaçamento destas moléculas, os polimorfos podem ser classificados como estáveis ou metaestáveis. A forma estável é a que possui em sua composição moléculas mais entrelaçadas umas com as outras, formando um retículo cristalino forte, e, portanto, com maior ponto de fusão, maior estabilidade termodinâmica e menor velocidade de dissolução do que as formas metaestáveis (14). Sob condições de temperatura e pressão definidas, apenas uma das possíveis formas polimórficas de uma substância pura é estável, sendo as outras formas denominadas metastáveis (16).
Se a qualquer temperatura e pressão uma das estruturas polimórficas é sempre a mais estável têm-se polimorfos monotrópicos, ou seja, a forma metaestável pode se converter na estável, mas o inverso não ocorre. Contudo, algumas substâncias apresentam polimorfos enantiotrópicos, o que significa que, sob diferentes condições de temperatura e pressão, a substância pode apresentar mais de uma forma estável, ou seja, existe uma temperatura de transição em que a estabilidade relativa se inverte, sendo possível obter qualquer dos polimorfos em função da temperatura (8; 17 - 19). Esta relação enantiotrópica, no entanto, é menos usual (14).
Os sólidos cristalinos também podem apresentar polimorfos a partir da inclusão ou incorporação de moléculas de solvente em suas redes cristalinas, sendo então conhecidos como solvatos, pseudopolimorfos, ou solvatomorfos (5), e se o solvente for a água, são denominados hidratos (13; 15; 20). Os solvatos podem conter quantidades estequiométricas ou não-estequiométricas de um solvente (13; 15; 20), que podem ser incoporados no cristal durante os processos de síntese (5), cristalização, liofilização, granulação, secagem ou armazenamento, por exemplo, gerando assim forças intermoleculares e estados energéticos distintos da forma cristalina pura (18), e, portanto, formas cristalinas com características físico-químicas e farmacêuticas distintas (15; 18; 20).
Diversos tipos de solventes orgânicos, como alcoóis, aromáticos, ésteres, éteres, e cetonas podem formar solvatos, sendo a N, N dimetilformamida (DMF), o dimetilsulfóxido (DMSO) e o 1,4-dioxano, solventes orgânicos com grande probabilidade de formar solvatos, provavelmente pela capacidade para formar ligações de hidrogênio (5).
Os solvatos também podem perder as moléculas de solventes incorporadas em seus cristais, processo conhecido como dessolvatação, e gerar formas menos ordenadas que suas formas cristalinas puras, mas mantendo a estrutura cristalina do solvato (15; 20; 21). Neste caso, estas formas são mais difíceis de caracterizar, pois os estudos analíticos indicarão que se tratam de formas cristalinas anidras quando, na verdade, possuem a estrutura do solvato da qual derivaram (15; 20).
Impacto do Polimorfismo na Indústria Farmacêutica
Um grande número de compostos farmacêuticos apresentam diferentes estruturas cristalinas, figurando dentre esses cerca de 70% dos barbituratos, 60% das sulfonamidas e 23% dos esteróides (22). Além das inúmeras propriedades físicas e químicas afetadas por este fenômeno, a possibilidade de manipulação da bioeficácia dos fármacos por meio do polimorfismo oferece aos cientistas farmacêuticos oportunidades interessantes, principalmente, no que diz respeito ao desenvolvimento de novos fármacos (22).
Deste modo, o polimorfismo tem contribuído significativamente para a variabilidade em produtos com desempenho na indústria farmacêutica, e ainda continua como um desafio para os cientistas da área em produzir medicamentos de qualidade consistente (23). Neste contexto, dois polimorfos de um mesmo composto químico podem ser tão diferentes em estrutura cristalina e propriedades como dois compostos distintos, sendo que essas diferenças manifestam-se enquanto o fármaco está em estado sólido, ou seja, uma vez obtida uma solução dos polimorfos as diferentes formas não podem mais ser distinguidas (24; 25). Portanto, podem ser esperadas diferenças na ação do fármaco, em termos farmacológicos e terapêuticos devido à presença de polimorfos em formas farmacêuticas sólidas, assim como em suspensões líquidas (24; 26).
Um exemplo clássico é o palmitato de cloranfenicol (PCA), cuja forma metaestável, forma polimórfica B, tem uma bioatividade oito vezes maior que a forma polimórfica A do PCA, e esta se administrada em humanos pode causar efeitos adversos como aplasia medular causando grandes prejuízos ao paciente (27). Para evitar a comercialização de medicamentos com problema de eficácia, a Farmacopéia Americana (USP 30th) implementou um teste para quantificar a presença do polimorfo indesejado A, sendo que este polimorfo não pode exceder 10% do IFA (28).
Já para a carbamazepina são conhecidas diversas formas sólidas: cristais anidros, solvatos, co-cristais e o sólido amorfo. Mas o insumo farmacêutico se apresenta quase sempre como cristal anidro (polimorfo p-monoclínico, triclínico ou misturas de ambos). Foram relatadas mortes pela utilização de medicamentos com este fármaco no final dos anos 80 nos EUA (29; 30). Isto fez com que o FDA iniciasse estudos sobre o comportamento do estado sólido do fármaco, em conjunto com diversas universidades. O estudo concluiu que a presença de diferentes polimorfos anidros e/ou do solvato diidrato era a causa do problema no medicamento. Desde então, a USP adicionou o ensaio de Difração de Raios X de pó (DRX de pó) na monografia da carbamazepina, de forma a garantir que o IFA utilizado possua apenas o polimorfo p-monoclínico (31 - 32).
Outro fator importante é que o polimorfo metaestável tende a se transformar no polimorfo estável, porém menos energético (transição polimórfica), o que pode ocorrer em função do tempo, da temperatura de armazenamento e do tipo de processo empregado na redução do tamanho de partículas e na produção do medicamento (33). Estas conversões podem gerar diversos impactos, sobretudo na velocidade de dissolução e solubilidade do mesmo fármaco (34 - 35).
A princípio, utilizar o polimorfo mais estável ao invés da forma metaestável é a solução definitiva para resolver o problema de conversão. Mas mesmo neste caso, também é possível que ocorram transformações durante os processos farmacêuticos (granulação, secagem, compressão). Portanto, independente da forma sólida escolhida do fármaco, sempre se deve identificar as etapas críticas do processo produtivo e os controles necessários para garantir que as características do medicamento se mantenham íntegras até o término do seu prazo de validade (36).
Enquanto que a estabilidade química é avaliada corretamente nos desenvolvimentos, a estabilidade física é muitas vezes deixada de lado ou realizada de forma inadequada. Por isso, cabe ressaltar que a estabilidade física também é importante, tanto para o IFA como para o produto farmacêutico final. O estudo da estabilidade física deve ser bem planejado para contemplar os aspectos particulares da formulação e deve ser capaz de determinar possíveis alterações no estado sólido do fármaco em matrizes complexas. Apenas com uma avaliação consistente do estado sólido é possível determinar, com segurança, a relação entre o polimorfo “candidato” do fármaco, o processo proposto e a qualidade do produto final (37).
Sendo verificado tais diferenças, é importante que no momento da aquisição da matéria-prima farmacêutica se especifique qual forma polimórfica deve ser utilizada nas etapas de desenvolvimento e produção. Caso contrário, pode-se obter um produto ineficaz devido ao comprometimento da dissolução do fármaco e, conseqüentemente, de sua biodisponibilidade (37).
A biodisponibilidade é a quantidade de fármaco que passa inalterada pela corrente sanguínea para, posteriormente, fixar-se em seus receptores e exercer a ação farmacológica. Em linhas gerais, as concentrações plasmáticas no sangue dependem da velocidade de liberação das moléculas desde sua forma farmacêutica, da velocidade de dissolução e absorção. Como cada polimorfo, geralmente, apresenta propriedades físico-químicas características, é possível observar comportamentos distintos durante os processos de liberação, absorção, distribuição, metabolismo e eliminação do fármaco (37).
O caso do ritonavir ficou mundialmente conhecido, pois durante o seu desenvolvimento e a prematura fabricação, parecia existir somente uma estrutura cristalina, em fase monoclínica (38). Esta forma, conhecida agora como forma I, não foi suficientemente bioeficaz quando administrada oralmente na forma sólida, requerendo que o produto (Norvir®) fosse formulado em uma cápsula gelatinosa mole contendo o fármaco dissolvido em uma solução hidro-alcoólica. Dois anos após o lançamento, vários lotes de cápsulas de Norvir® começaram a falhar nas especificações de dissolução. A avaliação dos lotes que apresentavam falhas revelou que uma segunda forma cristalina do ritonavir (forma II) havia precipitado. Após todo o esforço e tempo gastos para identificar o problema, este lote, contendo a forma II, 50% menos solúvel que a forma I, foi afastada do mercado, até que os produtores (a indústria Farmacêutica Abbott®) definissem um novo processo de síntese do ritonavir, livre de polimorfos indesejáveis (39; 40).
Este incidente suscitou discussões a respeito da possibilidade de ocorrerem desvios de qualidade devido ao polimorfismo nas mais diversas formulações e exemplos como esses motivaram autoridades reguladoras de vários países, principalmente da América do Norte e da Europa, a emitir resoluções para melhorar o controle do polimorfismo e diminuir a possibilidade de ocorrerem alterações na forma sólida do fármaco (39; 40).
O corpo humano possui aminoácidos, peptídeos, proteínas, ácidos graxos, lipídeos, etc., que são constituídos por diversos grupos funcionais. Estes grupos funcionais são necessários para que o fármaco possa interagir com as estruturas de interesse e, conseqüentemente, ter atividade farmacológica. Mas estes grupos funcionais também podem produzir diferentes interações inter e intramoleculares, que ocorrem por intermédio de pontes de hidrogênio, efeitos de cargas eletrônicas (interações eletrostáticas), forças de Van der Waals e ligações de hidrogênio não clássicas. Essas interações inter e intramoleculares diferenciadas podem produzir polimorfos distintos.
No ritonavir, por exemplo, o polimorfo I possui 6 pontes de hidrogênio intermolecular enquanto que o polimorfo II possui 8. Estas diferenças nas interações entre as moléculas dos diferentes polimorfos de ritonavir ajuda a explicar a diferença de solubilidade das duas formas cristalinas (40).
Os desvios de qualidade que têm ocorrido nos medicamentos devido ao polimorfismo têm acelerado a adoção de novas normas sobre o tema no setor farmacêutico. Elas discutem a identificação dos polimorfos e as questões relacionadas com o impacto e o controle do polimorfismo (41).
Mas cabe ressaltar que a maioria das normas e outros documentos oficiais que foram elaborados restringem a discussão às formas sólidas (muitas vezes apenas as formas sólidas orais) e às suspensões. Por não contemplarem as diversas formas farmacêuticas, estes documentos deixam lacunas importantes. Lacunas estas que se forem eliminadas no futuro poderão diminuir a probabilidade de ocorrerem novos casos como o do ritonavir (42 - 43).
A Conferência Internacional de Harmonização emitiu documentos que visam melhorar o controle do polimorfismo nos medicamentos novos e já existentes. Entre eles, o guia conhecido como “Guidance on Specifications: Test Procedures and Acceptance Criteria for New Drug Substances and New Drug Products: Chemical Substances.” (44) discute diversas questões, entre elas o polimorfismo. Entre as árvores de decisão utilizadas neste documento para indicar os procedimentos a serem seguidos durante as investigações, a árvore n° 4 trata do tema polimorfismo: Investigação da necessidade de estabelecer critérios de aceitação para o polimorfismo em fármacos e medicamentos (42; 43).
Para a agência Food and Drugs Administration (FDA) dos EUA (45), as questões relacionadas ao polimorfismo devem estar contempladas tanto na fase clínica (Investigational New Drug – IND) como na fase de registro (New Drug Application – NDA) de medicamentos inovadores (22; 43).
A preocupação com o polimorfismo também existe para os medicamentos genéricos, tanto que o guia para a indústria “Abbreviated New Drug Applications (ANDAs): Pharmaceutical Solid Polymorphism (Chemistry, Manufacturing and Controls Information) – CDER/FDA July 2007” trata apenas deste tema. O documento possui uma árvore de decisão subdividida em três árvores menores, onde se discute a investigação do polimorfismo e a definição de uma especificação para o IFA e o medicamento (22).
A Agência Nacional de Vigilância Sanitária (ANVISA) do Brasil também possui documentações que o tema polimorfismo é abordado, seguindo as tendências internacionais, de acordo com a tabela 1.

Uma nova forma polimórfica pode ser considerada uma invenção, e se tiver uma significativa aplicabilidade industrial poderá ser patenteada, a exemplo do que ocorreu com a ranitidina, ritonavir, ampicilina, palmitato de cloranfenicol, celecoxib, novobiocina, griseofulvina, indometacina, etc. As formas sólidas que mostram vantagens adicionais, em termos das propriedades físico-técnicas e físico-químicas, e apresentem realce na bioeficácia, podem ser entendidas como um novo fármaco. As pesquisas envolvendo a descoberta de novas formas cristalinas de fármacos são importantes para o processo de patentes, ajudando companhias inovadoras a manter a propriedade intelectual de uma substância (22; 43).
Existem numerosos exemplos onde companhias inovadoras adquiriram patentes sobre uma particular forma polimórfica, que se estenderam além da expiração da patente da molécula básica. Em detrimento disto, há uma grande preocupação da indústria farmacêutica em exaurir todas as possibilidades de polimorfos de uma dada substância e, assim, patentear todos os polimorfos possíveis de maneira a proteger o seu produto. Sabe-se que, pelas leis de patentes atuais, é considerado como nova patente qualquer fármaco que apresente uma nova forma polimórfica ou esteja co-cristalizado com outra substância. Para a indústria farmacêutica detentora da patente de um princípio ativo, isso pode significar um prejuízo enorme caso o novo polimorfo mostre biodisponibilidade equivalente ao(s) polimorfo(s) original (is) de sua patente. Portanto, isso abre uma nova vertente para os pesquisadores da área farmacêutica: a busca por um polimorfo ou co-cristal de fármacos já conhecidos, apresentando bioequivalência farmacêutica ou até mesmo promovendo as características farmacocinéticas em relação aos polimorfos conhecidos (22; 43).
SELEÇÃO DA FORMA POLIMÓRFICA MAIS ADEQUADA PARA PRODUÇÃO DE MEDICAMENTOS
A determinação prévia das possíveis fases cristalinas de candidatos a fármaco compreende uma etapa de extrema importância para pesquisa e desenvolvimento de medicamentos (5;50), pois sua concepção bem sucedida requer um amplo conhecimento das propriedades do estado sólido de todos os materiais envolvidos na formulação, inclusive dos excipientes. Embora esta investigação não garanta que todos os polimorfos de uma substância sejam descobertos, permite a verificação das fases cristalinas potenciais e garante que a fase ideal seja criteriosamente selecionada, minimizando o risco de aparecimento de polimorfos indesejados tardiamente (5).
A escolha de um componente ativo em uma única forma polimórfica para aplicação farmacêutica deve ser realizada através da caracterização das formas existentes e avaliação de uma série de fatores, tais como: higroscopicidade, sensibilidade ao calor, pressão e cisalhamento, capacidade de processamento, estabilidade química e física e biodisponibilidade (5), destacando-se os dois últimos como critérios decisivos ( 5; 17). Somente com esses dados é que uma seleção racional de forma sólida final pode ser realizada (51). Contudo, em se tratando de formas polimórficas com propriedades físico-químicas semelhantes, vale salientar que esta seleção tem menor importância (3; 17). O maleato de enalapril, por exemplo, possui duas formas polimórficas com solubilidade e características de dissolução equivalentes, sendo indiferente em termos de biodisponibilidade o emprego de uma ou outra fase na formulação farmacêutica (3). Isto também se aplica a fármacos, como o ácido mefenâmico, que possuem polimorfos com solubilidades distintas, mas cuja diferença na absorção e concentração plasmática não é significativa (18).
Outro aspecto de grande relevância envolve a seleção de formas anidras ou solvatadas, tendo em vista que durante o processo de desenvolvimento de formas farmacêuticas os fármacos frequentemente são expostos a solventes orgânicos e/ou aquoso que podem levar à formação de solvatos e hidratos (8). Essas formas geralmente possuem diferentes propriedades farmacêuticas, tais como biodisponibilidade, estabilidade ( 8; 10) e propriedades mecânicas (8), que podem influenciar na formulação, processo produtivo e estabilidade dos produtos farmacêuticos sob várias condições de armazenamento (10). Portanto, a escolha dessas formas para o desenvolvimento farmacêutico deve estar relacionada às propriedades supracitadas (8).
A seguir serão discutidos os principais critérios levados em consideração na escolha da forma polimórfica mais adequada para o desenvolvimento farmacêutico.
Forma Estável versus Forma Metaestável e Amorfa
Forma estável
Os polimorfos termodinamicamente estáveis, em regra, são preferidos para o desenvolvimento de medicamentos devido a sua maior densidade de empacotamento cristalino, orientação otimizada das moléculas, ligações quer sejam de hidrogênios ou não rede cristalina (8), menor energia interna, e consequentemente maior estabilidade química e física em relação às formas metaestáveis. Além do mais, as formas amorfas em geral são quimicamente instáveis, possuem maior higroscopicidade e tendem a se estabilizar em um sólido cristalino (4; 17) devido à falta de uma rede cristalina tridimensional, maior volume livre e maior mobilidade molecular (52).
A estabilidade química é um critério de extrema importância no desenvolvimento de medicamentos, tendo em vista que fármacos quimicamente instáveis degradam-se mais facilmente, podendo ocasionar uma diminuição de sua eficácia ou gerar metabólitos tóxicos. Já a estabilidade física está frequentemente relacionada ao menor risco de conversão de fase, cujas transformações mais freqüentes incluem a amorfização, interconversão desidratação-hidratação, desolvatação e transformação polimórfica, que podem ocorrer devido às variações de umidade, pressão e temperatura, durante a produção e/ou armazenamento do produto (5). Como essas conversões podem gerar polimorfos com propriedades distintas e afetar a estabilidade química, propriedades mecânicas e solubilidade dos fármacos, comprometendo a absorção oral do medicamento, é preferível o emprego da forma polimórfica mais estável a fim de garantir a reprodutibilidade de eficácia do produto durante sua vida de prateleira, sob uma variedade de condições reais de armazenamento (52). Este emprego é particularmente importante no processo produtivo de suspensões, tendo em vista que a transformação polimórfica pode gerar cristais de tamanhos diversos, afetando a estabilidade da formulação final (18)
No entanto, mesmo diante da maior estabilidade química e física do polimorfo de menor energia, deve-se exercer um controle adequado durante o processo produtivo a fim de evitar uma possível conversão de fase para outras formas polimórficas (52).
Forma metaestável e amorfa
Embora a forma polimórfica estável seja preferencialmente escolhida para o desenvolvimento de drogas, pelas razões já citadas no tópico anterior, há situações que requerem o emprego de uma forma metaestável cristalina ou amorfa (Tabela 1) (4; 52). Dentre estas situações, a mais comum consiste no aumento da taxa de dissolução de um medicamento devido à baixa solubilidade aquosa do polimorfo de menor energia (52). Isto porque as formas metaestáveis e amorfas possuem solubilidade aquosa superior, sendo a maior distinção observada entre materiais amorfos e cristalinos. Enquanto a diferença de solubilidade entre as formas cristalinas é normalmente inferior cerca de dez vezes, a diferença entre materiais amorfos e cristalinos pode ser até centenas de vezes (53). Por este motivo, alguns medicamentos são produzidos preferencialmente com o fármaco na forma metaestável, caso do palmitato de cloranfenicol (53) torsemida (3), fenilbutazona (18) mebendazol (34) e norfloxacina (19) ou na forma amorfa, caso da novobiocina (17), cefuroxima axetil (3), tetraciclina e cefalexina (18). Vale salientar que essa escolha também é extremamente útil para as novas moléculas farmacêuticas em desenvolvimento, tendo em vista que a maior parte delas apresenta baixa solubilidade aquosa (54). Ademais, também é uma opção para promover o aumento da taxa de dissolução de um medicamento, a fim de trazer alívio rápido para sintomas agudos (52).
Por outro lado, a seleção de uma forma metaestável ou amorfa devido à baixa solubilidade aquosa do polimorfo de menor energia deve ser avaliada conforme o Sistema de Classificação Biofarmacêutico (SCB). Segundo este sistema, apenas medicamento com taxa e extensão da absorção limitada pela sua dissolução tem biodisponibilidade e bioequivalência mais susceptíveis a ser afetadas pelos polimorfos de diferentes solubilidades. Em contrapartida, verifica-se uma menor susceptibilidade quando a taxa e extensão da absorção são limitadas pela permeabilidade intestinal. Ademais, as diferenças de solubilidade dos polimorfos dificilmente afetam a biodisponibilidade e bioequivalência de medicamentos com taxa de dissolução rápida em relação ao esvaziamento gástrico e quando as formas polimórficas são suficientemente elevadas. Por este motivo, fármacos que apresentam alta solubilidade aquosa não requerem o estabelecimento de uma forma polimórfica ideal para aumento de biodisponibilidade, situação, por exemplo, do cloridrato de ranitidina, cloridrato de terazosina (3) e metropolol (35).
Além do aumento da taxa de dissolução, os outros critérios para seleção estão relacionados à instabilidade do polimorfo de menor energia devido à justaposição de grupos reativos em moléculas adjacentes na rede cristalina, à facilidade de produção e a motivo de negócios, mais especificamente, patentes (52; 55). No que diz respeito à facilidade de produção, é preferível usar excipientes e processamento adequados para melhorar as propriedades mecânicas do polimorfo mais estável ao invés de escolher essas formas menos estáveis (52). Esta abordagem será mais bem discutida em tópico específico.
Apesar da menor estabilidade química das formas metaestáveis e amorfas, seu desenvolvimento como produto é possível através de uma formulação bem planejada, utilizando excipientes e processos de produção adequados (8; 52). Para um planejamento adequado é necessário um conhecimento profundo das características e comportamento do polimorfo, um desenho apropriado do medicamento e uma metodologia minuciosa de controle (4) que garanta a ausência de risco significativo para o paciente, pois a conversão de fase durante o tempo de vida de prateleira poderá acarretar um aumento da variabilidade farmacocinética e uma diminuição da taxa de dissolução, da biodisponibilidade e do desempenho in vivo (52). O caso do ritonavir é um dos mais conhecidos, pois chegou a ser retirado do mercado em 1998 por apresentar alterações de dissolução em lotes de cápsulas de Novir®, advindas do aparecimento da forma polimórfica II, após dois anos de comercialização do medicamento na sua forma polimórfica I, forma termodinamicamente menos estável, mas com solubilidade superior à forma II (18; 40).
Solvatos versus Hidratos
Solvatos
O emprego de solvatos não é recomendado para o desenvolvimento de medicamentos devido ao risco de dessolvatação e liberação de vapores orgânicos como impurezas ou como elementos tóxicos da formulação (5; 14). No entanto, determinadas substâncias apresentam grande tendência a formar solvatos (5), como a espironolactona, que exibe seis polimorfos, sendo dois anidras e quatro solvatos (35), além do exemplo clássico do antibacteriano sulfatiazol, que possui mais de 100 solvatos descritos na literatura (5). No mercado também se encontram disponíveis alguns medicamentos produzidos com fármacos na forma de solvatos, a saber: darunavir etanolato (Prezista®), sulfato de indinavir etanolato (Crixivan®), varfarina sódica isopropanol (Coumadin®) (5) e hiclato de doxiciclina (6).
Nestas circunstâncias, para o desenvolvimento de medicamentos seguros é necessário utilizar solventes de Classe III como etanol, 2-propanol e acetona em virtude deles apresentarem baixo potencial de toxicidade (5; 6). Os níveis máximos desses solventes permitidos em medicamentos estão estabelecidos no Guia para solventes residuais do International Conference on Harmonization of Technical (56). Outro aspecto da formulação que também deve ser analisado é a interação das moléculas dos solventes com os excipientes e IFAs (8;52) a fim de evitar possíveis alterações em sua estabilidade e performance (8).
Além do mais, os solvatos podem exibir diferentes propriedades físico-químicas e mecânicas em relação as suas correspondentes formas não solvatadas, podendo exercer impactos positivos ou negativos na formulação farmacêutica. Os solvatos de beclometasona, por exemplo, são bastante eficientes para micronização e formulação de suspensão e aerossóis sprays nasais (57). No que se refere à absorção in vivo, cada forma podem apresentar um comportamento distinto (27). Enquanto os solvatos de monoetanol de prednisolona apresentam maior taxa de absorção do que sua fase anidra, o solvato de hemiacetona apresenta praticamente a mesma taxa (27). Já os solvatos de gliblenclamida obtidos com pentanol e tolueno apresentam maior solubilidade e velocidade de dissolução do que suas formas polimórficas não-solvatadas (52). Adicionalmente, os solvatos apresentam baixa solubilidade em seus solventes de cristalização (53;57) devido a grande afinidade que possuem com o solvente que foi incorporado na rede cristalina (21) e a cristalização espontânea de solvatos produzir cristais de menor energia livre do que suas correspondentes formas não solvatadas (57).
Hidratos
A formação de hidratos é um fenômeno frequente que ocorre com moléculas orgânicas devido à natureza onipresente da água no ambiente (5). Estima-se que aproximadamente um terço das substâncias farmacologicamente ativas é capaz de formar hidratos (10; 20). Isto porque o pequeno tamanho da molécula de água favorece sua incorporação nos espaços vazios da rede cristalina, e porque o caráter multidirecional das ligações hidrogênio também é ideal para estabilização das estruturas cristalinas de grande parte das moléculas (20).
Em virtude de muitos sólidos farmacêuticos entrarem em contato com água durante algumas etapas do processo produtivo, tais como cristalização, liofilização, granulação via úmida, revestimento (aqueous film-coating) e spray-drying, bem como durante o armazenamento (58) se faz necessário conhecer informações sobre as condições de estabilidade, particularmente umidade e temperatura, nas quais as fases são estáveis (5). Pois, semelhante aos polimorfos em geral, a estabilidade física de hidratos e formas anidras pode depender da umidade relativa do ar e/ou temperatura do ambiente (5; 8). Ademais, alguns sais farmacêuticos, como sódio e cloridrato, formam hidratos com mais freqüência do que os “não-sais” devido à propensão da água para se ligar a sítios iônicos. Por outro lado, os “não-sais” são mais propensos a formar solvatos e polimorfos (5).
A seleção de hidratos para o desenvolvimento de medicamentos é complexa e deve considerar uma série de fatores, quais sejam: solubilidade, perfil de dissolução, processabilidade, comportamento de desidratação-hidratação e estabilidade de estado sólido (5). Dentre estes fatores, vale salientar que cristais hidratados tendem a exibir um perfil de dissolução inferior à de suas formas não hidratadas (5), pois baseando-se na teoria termodinâmica da solubilidade, um solvato é sempre menos solúvel do que o mesmo composto na forma original em seu solvente de cristalização. Assim, hidratos são menos solúveis em água do que sua respectiva forma anidra. Entretanto, se o solvente de formação do solvato for miscível em água, este solvato será mais solúvel em água do que sua forma anidra. Por exemplo, o hidrato de cafeína possui solubilidade aquosa muito menor que a solubilidade da sua forma anidra, entretanto, o hidrato tem solubilidade alcoólica muito maior que a anidra (53).
Apesar do menor perfil de dissolução, os hidratos são formas sólidas preferidas para o desenvolvimento de alguns medicamentos, como é o caso do cefadroxil (um monohidrato), cloridrato de paroxetina (um hemidrato) (6), alendronato de sódio, amoxicilina, atorvastatina e pantoprazol (5).
FORMAS DE AVALIAÇÃO DE POLIMORFISMO APLICADAS À INDÚSTRIA FARMACÊUTICA
Principais Técnicas de Obtenção
O processo de síntese de fármacos apresenta alguns pontos críticos que favorecem a transição cristalina, tais como a cristalização, secagem, estocagem e micronização (5). Apesar de constituir um desses pontos, a cristalização é um processo extensamente utilizado pela indústria farmoquímica na purificação e separação dos compostos, e é este processo que define o grau de pureza química e as propriedades físicas da substância que está sendo isolada, tais como tamanho e forma da partícula, estrutura cristalina e grau de imperfeições na mesma (59).
Esse processo é realizado a partir da obtenção de uma solução supersaturada do composto e posterior variação de alguns parâmetros, tais como: concentração da amostra a partir da evaporação lenta do solvente, temperatura, pH e força iônica. Deste modo, determinadas variações podem proporcionar ambientes termodinamicamente favoráveis à formação de outra estrutura cristalina, visando o retorno ao seu estado de equilíbrio (60).
Segundo Purohit &Venugopalan (2009) (19), a obtenção de cristais inicia-se com a fase de nucleação, que consiste na formação dos primeiros agregados ordenados, seguida pela fase de crescimento. A etapa de nucleação é bastante critica, uma vez que esta é determinante para a formação da forma sólida estável ou metaestável. Em soluções supersaturadas, por exemplo, qualquer ponto de nucleação é susceptível de conduzir à formação de material cristalino, mas nem sempre para o polimorfo mais estável (61).
Além da taxa de nucleação, outro ponto crítico no controle desse processo é a taxa de crescimento do cristal. Neste contexto, deve-se levar em consideração a temperatura e a importância do papel de cada solvente e impureza presente no processo de cristalização, pois estes têm influência direta na formação da estrutura cristalina. Dependendo do sistema de solventes a ser utilizado, a diferença de estabilidade termodinâmica de um composto em relação aos outros permite que esses sejam separados, onde a forma mais solúvel e menos estável se cristaliza antes (62).
Segundo Wang e colaboradores (2005) (63) e Cunha (2008) (60), um artifício freqüentemente utilizado pela Indústria Farmacêutica é a utilização de um anti-solvente, técnica conhecida como cristalização por adição de anti-solvente, na qual visa aumentar a taxa de crescimento de fármacos com crescimento lento em solventes orgânicos. O método consiste em adicionar um solvente que não solubiliza a droga em questão (por exemplo, água para drogas de baixa solubilidade aquosa), à solução do fármaco em líquido orgânico. Deste modo, Wang e colaboradores (2004) (64) utilizaram polietilenoglicol (PEG-300) como alternativa de solvente ou anti-solvente em substituição aos solventes orgânicos voláteis comumente utilizados para realização da cristalização e obtenção de polimorfos. Segundo os autores, nas condições do estudo o PEG apresentou a mesma capacidade de solubilização que os solventes 2-propanol e metanol para os compostos ibuprofeno, ácido salicílico e aminoácidos, avaliando-se a cristalização das substâncias quanto ao tempo de indução de nucleação e taxa de crescimento dos cristais. Deste modo, os autores observaram que a precipitação do ácido salicílico e glutamico por anti-solvente a partir da solução aquosas de PEG300 ou metanol por adição de água indicou um maior tempo de indução de cristalização para soluções de PEG, principalmente devido a uma maior energia interfacial de cristalização. Embora as taxas de crescimento dos cristais também foram menores em soluções de PEG do que em soluções de álcool tanto para o ácido salicílico quanto para o ácido glutâmico, as taxas ainda são bastante razoáveis para outras operações práticas. Ainda pode-se observar as mesmas estruturas cristalinas dos ácido salicílico e ácido glutâmico quando cristalizadas por álcoois alifáticos e PEG.
Por outro lado, Chen e Nan (2011) (65) utilizaram surfactantes mistos de brometo de hexadecil azanium (trimetil) e dodecil sulfato de sódio como antisolvente para mediar a nucleação e crescimento de cristais de CaCO3. Diferentes formas polimorficas foram obtidas (vaterite e aragonita). Tendo em vista que a adsorção do surfactante na superfície da amostra é diferente para os outros agentes devido aos efeitos das interações eletrostáticas repulsivas e hidrofóbicas, a energia superficial da amostra foi alterada por adsorções diferentes desses surfactantes. Esta mudança induziu a diferença de metamorfose e morfologia das amostras.
Outro método clássico consiste no resfriamento de uma amostra fundida abaixo do seu ponto de fusão, tendo em vista que pode ocorrer transição entre as formas polimórficas no momento do resfriamento, uma vez que as moléculas podem arranjar-se de maneira diferente no espaço formando retículos cristalinos diferentes (66). A obtenção de supositórios e algumas formas farmacêuticas semi-sólidas, tanto de uso tópico quanto sistemas matriciais de uso oral são exemplos de cristalização mediante resfriamento (67)
Adicionalmente, existem inúmeros métodos específicos para a obtenção de polimorfos. Embora muitos deles sejam baseadas no princípio da cristalização, sua especificidade se dá pela junção desta técnica com novas ferramentas cada vez mais eficientes. Esses métodos reduzem a barreira de ativação para nucleação e, assim, aceleram o processo de cristalização, sendo particularmente úteis para substâncias que são difíceis de cristalizar (5; 61; 62; 68).
Neste contexto, Capes e Cameron 2007 (69) investigaram o efeito, das condições de evaporação lenta de solvente sobre a cristalização polimórfica do paracetamol e observaram a obtenção da forma cristalina metaestável do fármaco. Uma vez que os polimorfos metaestáveis apresentam alta energia interna e, conseqüentemente, apresentam maior velocidade de dissolução e biodisponibilidade do que os estáveis, o uso de polimorfos metaestáveis muitas vezes são requisitados como estratégia para o incremento de solubilidade de fármacos pouco hidrossolúveis. Embora exista o risco potencial de conversão para o polimorfo termodinamicamente mais estável ou para as outras possíveis formas polimórficas (59) outras técnicas podem ser utilizadas visando a estabilização da estrutura cristalina, evitando sua transição, como demonstra o xestudo de Capes & Cameron 2007 (69), no qual alcançou a estabilidade a partir da cristalização por superfície de contato.
A utilização da técnica de fluido supercritico têm sido aplicada para a fabricação de polimorfos puros. Esta técnica geralmente é realizada em um processo de etapa única, sendo possível isolar diferentes polimorfos por meio do controle da temperatura, pressão, vazão e solvente(68). Utilizando condições de alta pressão, Tozuka (2003) (70) obteve transformação física sólido-sólido de cristais do ácido deoxicólico (DCA) empregando dióxido de carbono supercrítico.
Apesar das diferentes técnicas de cristalização existentes, não há um método que possa fornecer confiança absoluta do isolamento de uma determinada forma polimórfica. Para tanto, dispõe-se de diversos métodos eficientes que auxiliam na elucidação do polimorfo obtido (61), como será discutido adiante.
Principais Técnicas de Identificação e Caracterização
A variação considerável nas propriedades entre os diferentes polimorfos, faz a caracterização de todas as formas polimórficas essencial para a completa compreensão do nível estrutural de qualquer material molecular (3). Dessa forma, os polimorfos podem ser caracterizados e diferenciados pelas suas propriedades físico-químicas empregando-se técnicas analíticas como: difração de raios X, análise térmica, microscopias e espectroscopias (44).
Difração de raios-X (DRX)
A DRX é a técnica mais amplamente utilizada na caracterização microestrutural de materiais cristalinos. Investiga e distingui diferentes formas cristalinas de substâncias (71)
Quando o feixe de raio X incide sobre o material, os mesmos são espalhados elasticamente sem perda de energia pelos elétrons dos átomos, se os átomos estiverem arranjados de maneira sistemática como uma estrutura cristalina, o espalhamento torna-se periódico, formando um padrão de difração característico determinado pela estrutura cristalina do sólido, se o material for amorfo, não apresenta esse padrão de difração, portanto é um método rápido e eficiente para avaliar se uma amostra se encontra no estado amorfo ou cristalino (72)
Permite também identificar as fases cristalinas, presentes numa amostra, sendo necessário a obtenção do monocristal para a realização do ensaio de difração de raios-X de monocristal, e as vezes é o único meio de determinar, entre os possíveis polimorfos de uma substância, a forma estrutural e conformação predominante, bem como as interações intermoleculares (73;74).
No entanto, uma vez que apenas um polimorfo é termodinamicamente estável sob um dado conjunto de condições ambientais, muitas vezes é impossível obter cristais de tamanho adequado e qualidade para a análise de estrutura simples de um único cristal. Neste caso, deve-se recorrer a técnica de DRX pó onde a sobreposição de reflexões limita as informações. Entretanto pelo alto custo do equipamento, o uso da técnica torna-se inviável para muitos laboratórios de pesquisa e indústrias (67).
A cristalografia de raios-X é, naturalmente, a técnica de escolha para a identificação definitiva e direta de diferentes polimorfos. No entanto, nem sempre é aplicável, no caso para a a coexistência de diferentes polimorfos no mesmo cristal (75).
Desta forma, a caracterização de polimorfos é feito, em geral, utilizando-se outras técnicas analíticas que, em conjunto, geram dados capazes de confirmar a presença de diferentes formas cristalinas.
Espectrometria na região do infravermelho (IV)
As análises por espectrofotometria de infravermelho são essenciais na caracterização de estruturas polimórficas, quanto a sua estrutura química indicando se houve ou não modificação no espectro de absorção das bandas dos principais grupamentos funcionais, após o processo de cristalização (76).
Girlando e colaboradores (2010) (75) identificaram dois polimorfos, o R e β-TPB e perceberam diferenças claras entre os espectros IV das duas fases, confirmando que a conformação molecular é diferente.
Lohani e colaboradores 2011 (77) utilizaram técnicas espectroscopicas para verificar a agregação molecular da forma polimorfica I da Indometacina em soluções de acetonitrila e etanol. Seus dados evidenciam a formação de interações intermoleculares (ligações de hidrogênio) entre a água residual do soluto e solvente. Perceberam que os efeitos das interações são importantes para determinar o inicio da nucleação nas soluções supersaturadas que é um factor determinante da taxa relativa de nucleação e crescimento de cristais de polimorfos, e assim pôde controlar a cristalização preferencial da forma polimórfica I.
Espectroscopia de espalhamento Raman
A espectroscopia Raman permite uma caracterização mais facil das diferenças estruturais do polimorfo , pois devido a habilidade de obter espectros em frequencias vibracionais mais baixas , pode dar informaçoes vibracionais sobre a rede de um cristal (78).
A técnica fornece informações quimicas similares a IV, porém complementares, uma vez que a absorção no IV depende da variação do momneto dipolo da molecula e o Raman da variação da polarizabilidade, entretanto quando aplicado a identificação de polimorfos, a tecnica possui diversas vantagens. É batante adequada para aos estudos in situ, podendo ser realizada mesmo na presença de agua ou atraves do vidro, eliminado a necessidade de preparação da amostra (79).
Devido a sua facilidade e a atual exigencia da legislação pelo controle das formas polimórficas, a busca pela aplicação da tecnica vem crescendo e se ampliando (80).
A espectroscopia Raman é bastante utilizada nos ensaios de controle de qualidade de medicamentos. O método é perfeitamente rápido e adequado para satisfazer as exigências regulamentares de monitoramento de formas de cristal durante o processamento e armazenamento e, muitas vezes consegue detectar a forma de cristal presente em medicamentos mesmo quando os excipientes utilizados não são conhecidos (81).
Ressonância magnética nuclear de hidrogênio (RMN’H)
O uso da RMN na investigação de polimorfismo é facilmente entendido, pois se um composto apresenta dois polimorfos, a e b, suas formas cristalinas são conformacionalmente diferentes. Isto significa que um determinado carbono presente na forma a pode possuir uma geometria molecular ligeiramente diferente quando comparada com o mesmo carbono na forma b, o que pode originar ambientes locais distintos, apesar de possuírem os mesmos átomos ligados entre si. A diferença no ambiente local pode acarretar diferentes interações de deslocamento químico para o mesmo átomo de carbono, nas duas diferentes formas polimórficas (82).
Assim, os parâmetros do RMN podem ser correlacionados com características estruturais, tais como subunidades local, distâncias de ligação e ângulos, ou geometrias. Além disso, as intensidades dos sinais de RMN são proporcional ao número de átomos por célula unitária, e pode ser determinada a ocupação relativa de diferentes sítios atômicos, bem como idetificar a quantidade de polimorfos existentes (83).
Uma das principais vantagens da RMN é a possibilidade de utilização desta técnica no produto acabado, ou seja, no fármaco formulado, pois o deslocamento químico do polimorfo é sensível ao ambiente químico e à conformação molecular, o que permite investigar a conversão nos vários polimorfos durante o processamento (82).
Shaibat e colaboradores 2007 (84) discutiram a possibilidade de distinguir polimorfos de complexos de Cu (II) paramagnético por RMN’H e C13 e demonstrram que espectros para polimorfos de complexos paramagnéticos apresentam diferenças extraordinariamente grandes em larguras de linha e posições espectrais.
Microscopia eletrônica de varredura (MEV)
Esta técnica é muito útil para o estudo estrutural externo da amostra, no caso dos polimorfos é importante para a caracetrização dos efeitos superficiais (59).
Sheikhzadeh e colaboradores (2006) (85) caracterizaram a forma 1 e 2 de cloridrato de buspirona, um medicamento ansiolítico. As técnicas utilizadas para a caracterização incluiu microscopia, análise térmica, infravermelho, difração de raios-X e espectroscopia Raman. Morfologicamente, a forma 1 e 2 consistem de placa e cristais colunares, respectivamente.
Em meados de 1998, vários lotes de cápsulas de ritonavir tiveram diferenças no estudo de dissolução e foram examinados usando microscopia e difração de raios X, um novo polimorfo foi identificado, por isso que havia reduzido a solubilidade comparado com a forma de cristal original (40).
Através das fotomicrografias obtidas pelo MEV para caracterizar ou identificar diferentes formas polimórficas, pode-se observar diferenças significativas na morfologia das estruturas.
Analise térmica
Analise térmica é o termo usado para descrever técnicas analíticas que medem as propriedades físico-químicas das amostras em função da temperatura ou o tempo quando a amostra é submetida a uma variação de temperatura controlada. Entre as diversas técnicas termoanalíticas a Termogravimetria (TG) e Calorimetria Exploratória Diferencial (DSC) são as mais utilizadas na área farmacêutica na caracteriza¬ção térmica de fármacos, na determinação de pureza, em estudos de compatibilidade de formulações farmacêuticas, na identificação de polimorfos, na avaliação de estabilidade e em estudos de decompo-sição térmica de fármacos e medicamentos (86; 87).
Quanto à avaliação de presença de polimorfismo nos fármacos por DSC, o resultado é o aparecimento de múltiplos picos de fusão relacionados a cada forma cristalina, a velocidade de aquecimento é crítica neste ponto. Um aquecimento muito rápido pode ocultar uma curva endotérmica, enquanto um aquecimento muito lento pode permitir uma transição ou decomposição, dificultando a visualização dos eventos térmicos. Por este motivo, a pesquisa de cristais deve ser executada, inicialmente, por meio de um estudo em duas razões de aquecimento, 2 e 20 ºC/min. (16; 88).
Outros aspectos importantes para a observação de polimorfos na curva DSC são: transição cristalina que ocorre antes do ponto de fusão do polimorfo de maior ponto de fusão, em que o pico será endotérmico para polimorfos enantiotrópicos (processo reversível) e exotérmico para polimorfos monotrópicos (processo irreversível) e nenhuma perda de massa é observada na curva TG, e o processo de recristalização, representado por um pico exotérmico que ocorre após a fusão da forma de menor ponto de fusão (89; 90).
Oliveira e colaboradores (2010) (91) promoveram a cristalização da sinvastatina em diferentes solvente e verificaram a cristalização do fármaco em duas formas acicular e tabular. Com o objetivo de identificar as formas polimórficas, curvas de DSC foram obtidas para as formas acicular e tabular, e não houve duplicidade de pontos de fusão e nem aparecimento de eventos de transição cristalina, indicando não haver polimorfismo. Ainda assim, submeteram os difratogramas experimentais ao ajuste pelo método de Rietveld, e a estrutura prevista para ambos os difratogra¬mas é a de um cristal de forma tabular para a sinvastatina. Diante disto, a sinvastatina não apresenta o polimorfismo clássico, mas apresenta um polimorfismo do tipo morfológico, onde o fármaco tem a mesma cela unitária, mas com hábitos cristalinos diferentes devido a interações soluto-solventes preferenciais no processo de crescimento dos cristais. Este polimorfismo morfológico não altera as propriedades físico-químicas e, desta forma, ambas as formas cristalinas, acicular ou tabular, apresentam a mesma estabilidade, solubilidade e reatividade.
Diante deste fato, esse tipo de polimorfismo onde obtem-se cristais com morfologias diferentes, mas com mesma cela unitária, podendo este fato acontecer por influência no eixo de crescimento do núcleo, que é dependente de concentração, temperatura e tipo de solventes utilizados, só poderá ser identificado por Microscopia Eletronica de Varredura, em curvas DSC não são identificados múltiplos picos de fusão, enquanto na análise por DRX não é possível observar diferenças cristalográficas entre as formas cristalinas. No entanto, no DRX se observam diferenças na intensidade, em planos específicos de reflexão, o que sugere cristais de mesma cela unitária com orientação preferencial para o desenvolvimento macroscópico dos cristalitos de formas diferentes (16).
Silva e colaboradores (2009) (92) utilizaram a técnica de DSC-photovisual para identificar e caracterizar a qualidade de sulfato indinavir em amostras comerciais de acordo com suas propriedades termoanalíticas, é um método que permite o acompanhamento das altera¬ções produzidas numa amostra por variação de temperatura através de observação microscópica com luz polarizada. Este método é de grande utilidade e é complementar ao DSC para informações de compatibilidade de formulações e identificação de polimorfismo. Sendo assim, o DSC-photovisual demonstrou a formação de dois tipos de solvatos com mudanças no pico de fusão e ponto de fusão do sulfato de indinavir. A presença de diferentes quantidades de água na molécula de indinavir pode alterar as propriedades térmicas e físico-químicas (por exemplo, solubilidade e biodisponibilidade) do fármaco.
Dissolução
A presença de diferentes polimorfos em uma formulação pode comprometer a dissolução de um fármaco a partir de sua forma farmacêutica, uma vez que os polimorfos freqüentemente apresentam diferentes solubilidade. Em alguns casos a diferença não é significativa. Mas quando ocorre o oposto, os medicamentos podem se tornar menos ativos, inativos ou tóxicos quando existe uma relação direta entre esse parâmetro e a atividade farmacológica. O efeito do polimorfismo na biodisponibilidade é a conseqüência mais importante para os fármacos, se a biodisponibilidade é mediada através da dissolução. (34; 67)
O ensaio de dissolução do medicamento determina a massa dissolvida do fármaco em função do tempo, enquanto que a dissolução intrínseca determina a velocidade de massa dissolvida do fármaco. Por isso, quando se avalia os diferentes polimorfos nestes testes, eles normalmente se tornam mais eficientes que a solubilidade para predizer o desempenho dos diferentes polimorfos em uma formulação. Um dos primeiros exemplos apresentados na literatura, que mostrou mudanças significativas na biodisponibilidade devido ao polimorfismo foi o Palmitato de clorafenicol. A sua forma B tem uma atividade terapêutica oito vezes maior que a forma A (80). Este incidente suscitou discussões a respeito da possibilidade de ocorrerem desvios de qualidade devido ao polimorfismo nas mais diversas formulações.
Froehlich e colaboradores (34) utilizaram a espectroscopia de infravermelho e o perfil de dissolução in vitro para caracterizar matérias-primas e comprimidos (referência e genéricos) existentes no mercado brasileiro. Os resultados obtidos demonstram que as três formas polimórficas do mebendazol estão presentes nos medicamentos e duas delas em matérias-primas, sugerindo que um maior controle deveria ser utilizado para a seleção de matérias-primas que apresentam polimorfismo, assegurando, através de testes simples e rápidos, a qualidade de medicamentos genéricos.
Exemplos como esses motivaram autoridades reguladoras de vários países, principalmente da América do Norte e da Europa, a emitir resoluções para melhorar o controle do polimorfismo e diminuir a possibilidade de correrem alterações na forma sólida do fármaco (59).

INTERFERÊNCIA DO POLIMORFISMO NO PROCESSAMENTO INDUSTRIAL DE MEDICAMENTOS
Segundo Szterner e colaboradores (2010) (94), medicamentos recém-elaborados ou já consagrados podem apresentar alterações imprevistas em termos de solubilidade aquosa dos componentes e estabilidade do medicamento, repercutindo diretamente na sua biodisponibilidade, e tornando-se passível, portanto, do comprometimento da sua eficácia e segurança.
Uma vez que variações no hábito cristalino das partículas podem acarretar consideráveis variações nas suas propriedades físico-químicas, estas constituem uma das maiores problemáticas da Indústria Farmacêutica. Neste contexto, conhecer todas as possíveis formas cristalinas do material torna-se imprescindível para o planejamento racional do desenvolvimento da forma farmacêutica, permitindo assim o desenho de um processo de fabricação eficiente para a forma cristalina empregada (95), visando prever os possíveis problemas de produção e efeito biológico, e gerar artifícios para contorná-los.
Portanto, determinar a estrutura cristalina de um composto ativo através dos métodos de caracterização é uma das primeiras etapas do desenvolvimento farmacêutico, uma vez que esta pode sinalizar o quão facilmente ele pode ser formulado (50). Variações na estrutura cristalina dos pós dos diferentes polimorfos podem acarretar mudanças nas suas propriedades tecnológicas, como plasticidade, tamanho e morfologia de partícula e, consequentemente, propriedades de fluxo, as quais afetam diretamente o processo de fabricação do medicamento (95; 52).
Caracterização: delineamento do processo de produção
Segundo Nery e colaboradores (2008) (96), partículas esféricas apresentam comportamento de compressão e fluxo previsível por modelos relativamente simples. Entretanto, desvios da forma esférica ou, formas difíceis de serem descritas por figuras geométricas simples, possuem desempenho produtivo mais difícil de ser previsto. Ademais, a estrutura cristalina mais estável de determinada molécula apresenta forças intermoleculares mais fortes, ampliando sua densidade de empacotamento. Tal característica repercute na redução da sua capacidade de deformação, de maneira que, após o processo de compressão, obtêm-se comprimidos com maior grau de porosidade em comparação às formas metaestáveis, já que estas últimas possuem menor intensidade das forças intermoleculares e, conseqüentemente, maior facilidade de compressão (97).
Por este motivo, sólidos amorfos apresentam melhores propriedades mecânicas em relação aos sólidos altamente cristalinos, uma vez que consistem em estruturas desorganizadas e, por isso, possuem forças intermoleculares muito fracas, proporcionando-lhes grande capacidade de compressão. Adicionalmente, possuem outros atributos favoráveis no campo farmacêutico, uma vez que apresentam alta hidrossolubilidade e taxa de dissolução (20).
Em contrapartida, algumas complicações de formulação e comportamento biológico podem ser observadas devido à desorganização molecular que envolve estas partículas, uma vez que são termodinamicamente instáveis e, portanto, susceptíveis a modificação estrutural, tendendo à estabilidade (98). Uma vez que pequenas modificações na densidade de empacotamento da partícula ocasionam alterações significativas na estrutura cristalina e, consequentemente, solubilidade e estabilidade, a utilização de partículas amorfas torna-se questionável (99).
Deste modo, formas cristalinas estáveis tendem a ser preferidas em relação às formas metaestáveis e amorfas (53; 18). Porém, a estabilidade destas estruturas geralmente confere propriedades mecânicas inadequadas, além de limitações em termos de biodisponibilidade. Neste contexto, torna-se imperioso a intervenção da tecnologia farmacêutica visando à escolha de excipientes e processos adequados para a forma cristalina em questão.
Entretanto, uma forma cristalina metaestável pode ser suficientemente estável nas condições ambientais, tornando-se, portanto, uma excelente alternativa de incremento de solubilidade aquosa e adequação de suas propriedades físicas aos processos envolvidos. Este fenômeno pode ser exemplificado com o grafite e o diamante que, embora sejam constituídos da mesma estrutura molecular, a forma estável (diamante) necessita de alta energia de ativação e, por isso, o grafite (forma metaestável) é o mais encontrado na natureza (59). Por este motivo, técnicas de cristalização são frequentemente empregadas a fim de induzir a obtenção de cristais com morfologia específica, visando aperfeiçoar o processo de produção e contornar possíveis problemas de biodisponibilidade. Segundo Shekunov & York (2000) (100) este artifício foi utilizado no desenvolvimento de diversos medicamentos a partir de fármacos como nitrofurantoína, ibuprofeno, octotiamina e acetominofeno.
Por outro lado, nas situações em que as formas metaestáveis e amorfas são requisitadas e estas não apresentam estabilidade suficiente, deve-se também lançar mão da tecnologia farmacêutica visando, no entanto, a estabilização do fármaco, a fim de evitar possíveis conversões de fase ao longo de todo o processo produtivo e período de armazenamento do medicamento (101).
Portanto, a seguir serão abordados os pontos do processo produtivo de medicamentos que podem proporcionar modificações na forma cristalina empregada, tanto do insumo farmacêutico ativo quanto dos excipientes, podendo agregar valores negativos ao produto no que se refere a sua eficácia e segurança terapêutica. Além do mais, também serão discutidos os pontos que podem levar a modificações intencionais que tragam benefícios ao processo e/ou ao produto acabado.

Translation - English POLYMORPHISM: CHALLENGES AND RESEARCH STRATEGIES, DEVELOPMENT AND INNOVATION IN THE PHARMACEUTICAL INDUSTRY
Polymorphism of active pharmaceutical ingredients (APIs) and excipients comprise a huge challenge for research, development and innovation (R,D & I) in the pharmaceutical industry, because solid state properties are directly related to crystalline structure of molecules. Thus, the use of different polymorphic forms of the same compound can generate a direct impact on the formulation and production of medicines and where the mechanical properties, the bioavailability, and the stability of the most critical points are evaluated in product development. In this context, this paper aims to discuss, based on a review of the literature, the main aspects affected by polymorphism, as well as strategies and methods for characterization, collection and selection of polymorphic phases most suitable for the production of medicines. Moreover, decisive factors will be considered in choosing the most suitable processes and excipients for the control of polymorphism during production and storage of pharmaceuticals.
INTRODUCTION
The ability of a solid to exist in more than one crystalline structure from the same molecular structure, a phenomenon known as polymorphism, comprises an area of great interest and concern for the pharmaceutical industry (1). This is because different active pharmaceutical ingredients (APIs) have different polymorphic forms (2), which have quite distinct physical-chemical, thermodynamic, kinetic, interfacial and mechanical properties (3-5). Some of these properties such as solubility and stability are essential for successful development of drug candidates (6) and therefore these differences could generate a direct impact both in performance and stability of the pharmaceutical product, but also in the processing capacity of the API and the design and production of the drug (3,7).
The crystalization of active pharmaceutical ingredients (APIs) particularly those that possess multiple polymorphic forms, is among the most critical processes and least understood of pharmaceutical production. A number of failures in the processes and products can be attributed to an insufficient understanding and control of crystallization processes (8). Thus, the unexpected appearance or disappearance of a polymorphic form, for example, can lead to serious consequences which may result in the delay of the development of the drug and / or the interruption of commercial production (3). A decisive episode, which occurred in 1998, was the temporary removal of the drug Norvir®, due to the appearance of a previously unknown polymorph, Ritonavir (6).
As a result, systematic procedures of polymorphic characterization must be still conducted in stages of preformulation. (9) Therefore, understanding the relationship between solid state properties and crystal structures of possible polymorphic phases of an API (10) allows for the choice of the most appropriate form of development of a medicine (9) and can also be used to improve operational strategies and formulation as well as improve the design of suitable protocols for stability (10) in order to avoid problems during or after the launch of a product (11).
Furthermore, control of polymorphism has become one of the key issues with regard to scientific research related to the pharmaceutical industry (7) since most products are formulated with pharmaceutical excipients and drugs in the solid state and crystalline form (5, 7 , 12). The development of these molecular crystals in solid pharmaceutical forms is preferred for reasons of production, stability (5), convenience of administration and dosage for the patient (4), even though there are also challenges, such as polymorphism (5).
Thus, this paper aims to discuss, based on of a review of the literature, the challenges posed by polymorphism in research, development and innovation of drugs and the strategies and methods employed for the characterization, collection and selection of polymorphic phases as well as the choice of the most appropriate procedures and excipients for the control of polymorphism during production and storage of medications.
Types of Polymorphism
Substances in the solid state may be in the amorphous form of molecules with tangles and without a defined crystalline network (13) or crystalline, with molecules arranged in a repeating pattern ordered throughout the particle (14). These crystalline substances can be formed by flexible molecules that adopt a more flexible solid state conformation (conformational polymorphism), or rigid molecules that take different forms of packaging (packaging polymorphism ) (2, 5, 15).
Depending on the interlacement of these molecules, the polymorph can be classified as stable or metastable. The stable form is that which has in its composition molecules intertwined with each other, forming a strong crystalline lattice, and, therefore, a higher melting point, greater thermodynamic stability and a lower dissolution rate than the metastable form (14) . Under set conditions of temperature and pressure , only one of the possible polymorphic forms of a pure substance is stable and the others metastable forms (16).
If at any temperature and pressure one of the polymorphic structures is always the most stable then you have monotropic polymorphs, i.e., the metastable form can become the stable, but the converse does not occur. However, certain substances have enantiotropic polymorphs, which means that, under different conditions of temperature and pressure, the substance may have a more stable form, i.e., there is a transition temperature where relative stability is inverted, and any of the polymorphs may be obtained according to the temperature (8, 17-19). This enantiotropic relationship, however, is less usual.(14).
Crystaline solids may also have polymorphs from the inclusion or incorporation of solvent molecules in their crystalline lattices and is then known as solvates, pseudo-polymorphs or solvatomorphs (5) and if the solvent is water, they are called hydrates (13; 15, 20). The solvates may contain stoichiometric or non-stoichiometric amounts of a solvent (13, 15, 20) which can be incorporated into the crystal during synthesis processes (5), crystallization, lyophilization, granulation, drying or storage, for example, thus generating intermolecular forces and different energy states of the pure crystalline form (18), and therefore, crystalline forms with physical-chemical and distinct pharmaceuticals (15, 18, 20).
Various types of organic solvents such as alcohols, aromatics , esters, ethers, ketones can form solvates, where N, N-dimethylformamide (DMF), dimethylsulfoxide (DMSO), and 1,4-dioxane, are organic solvents with a high probability of forming solvates, probably due to the ability to form hydrogen bonds (5).
The solvates may also lose molecules of solvents incorporated in its crystals, the process known as desolvation, and generate forms less ordered than its pure crystalline forms, but maintaining the crystalline structure of the solvate (15, 20, 21). In this case, these forms are more difficult to characterize, for analytical studies indicate that these are anhydrous crystalline forms when, in fact, they have the structure of the solvate from which they are derived (15, 20).
Impact of Polymorphism on the Pharmaceutical Industry
A large number of pharmaceutical compounds exhibit different crystalline structures, among those about 70% are of barbiturates, 60% sulfonamides and 23% steroids (22). Beyond the numerous chemical and physical properties affected by this phenomenon, the possibility of manipulation of the bio-effectiveness of drugs by means of polymorphism offers pharmaceutical scientists exciting opportunities, especially as regards to the development of new drugs (22).
Thus, polymorphism has contributed significantly to the variability in products with performance in the pharmaceutical industry, and still remains a challenge for scientists in the field to produce drugs of consistent quality (23). In this context, two polymorphs of a same chemical compound may have such different crystalline structures and properties as two distinct compounds, such that these differences manifest themselves while the drug is in the solid state, i.e., once a polymorph solution is obtained, the different forms cannot be distinguished. (24; 25) Therefore, differences can be expected in the drug, in pharmacological and therapeutic terms due to the presence of polymorphs in solid dosage forms, as well as liquid suspensions (24, 26).
A classic example is chloramphenicol palmitate (CPE), whose metastable form, B polymorphic form, has a bioactivity eight times higher than the A polymorphic form of CPE, and this if administered in humans can cause adverse effects such as bone marrow aplasia causing great harm to the patient (27). To avoid the commericialization of drugs with problems of efficacy, the United States Pharmacopeia (USP 30th) implemented a test to quantify the presence of the undesired polymorph A, provided that this polymorph cannot exceed 10% of API (28).
As for carbamazepin, various solid forms are known: anhydrous crystals, solvates, co-crystals and the amorphous solid. But the pharmaceutical ingredient is usually present as anhydrous crystal (p-monoclinic polymorph, triclinic or mixtures of both). Deaths were reported by the use of medicines with this drug in the late 80s in the USA (29; 30). This led the FDA to initiate studies on the behavior of the solid drug, together with several universities. The study concluded that the presence of different anhydride polymorphs and / or dihydrate solvates was the cause of the problem in the medicine. Since then, the USP added the diffraction X-ray powder test (DXR powder ) in the carbamazepine monograph, to ensure that the API has only used the p-monoclinic polymorph (31-32).
Another important factor is that the metastable polymorph tends to become the stable polymorph but less energetic (polymorphic transition), which can occur as time dependent on the storage temperature and the type of process employed to reduce particle size and in production of the medicine (33).
These conversions may generate various effects, especially in dissolution rate and solubility of this same drug (34-35).At first, to use the more stable polymorph rather than the metastable form is the ultimate solution to solve the conversion problem. But even in this case, it is also possible that changes will occur during the pharmaceutical processes (granulation, drying, compression). Therefore, regardless of the solid form of the drug chosen, one should always identify the critical stages of the production process and the necessary controls to ensure that the product characteristics remain intact until the end of its validity (36).
While chemical stability is evaluated correctly in development, physical stability is often left out or improperly performed. Therefore, it is noteworthy that physical stability is also important for both the API as for the final pharmaceutical product. The study of physical stability must be planned to contemplate the particular aspects of the formulation and must be able to determine possible changes in the solid state of the drug in complex matrices. Only with a consistent evaluation of the solid state can it be determined with certainty, the relationship between the "candidate" polymorph drug, the proposed process and the final quality of the product (37).
With such differences found, it is important that upon the acquisition of the pharmaceutical raw material that it is specified which polymorphic form should be used in the stages of development and production. Otherwise, you can get an ineffective product due to the impairment of the drug dissolution and, consequently their bioavailability (37).
Bioavailability is the amount of drug which passes unchanged through the bloodstream to later attach itself to their receptors and exert the pharmacological action. In general, plasma concentrations in the blood depend on the speed of release of molecules from its dosage form, dissolution and absorption rate. As each polymorph usually has characteristic physical-chemical properties, it is possible to observe distinct behavior during the release, absorption, distribution, metabolism and elimination processes of the drug (37).
The case of ritonavir became known worldwide, because during its early development and manufacturing, there appeared to be only one crystalline structure in the monoclinic phase (38). This form, now known as Form I, was not sufficiently bioeffective when administered orally in solid form, requiring that the product (Norvir®) be formulated into a soft gelatin capsule containing the dissolved drug in a hydro-alcohol solution. Two years after launch, several lots of capsules of Norvir® began to fail in the dissolution specifications. The evaluation of the lots which had failures revealed that a second crystalline form of ritonavir (Form II) had precipitated. After all the time and effort spent to identify the problem, this lot, containing Form II, 50% less soluble than Form I, was taken off the market until producers (Abbott® pharmaceuticals industry) could define a new synthesizing process for ritonavir, free of undesirable polymorphs (39, 40).
This incident suscitated discussions about the possibility of occurrence of quality deviations due to polymorphism in various formulations and examples such as these led regulators in several countries, especially North America and Europe, to deliver resolutions to improve the control of polymorphism and reduce the possibility of occurrence of changes in the solid form of the drug (39, 40).
The human body posseses amino acids, peptides, proteins, fatty acids, lipids, etc., which consist of diverse functional groups. These functional groups are necessary so the drug can interact with the structures of interest and, consequently, have pharmacological activity. But these functional groups can also produce different inter- and intramolecular interactions that occur through hydrogen bridges, effects of electronic charges (electrostatic interactions), Van der Waals forces and non-classical hydrogen bonding. These differentiated inter-and intramolecular interactions can produce distinct polymorphs.
In ritonavir, for example, polymorph I posseses 6 intermolecular hydrogen bridges while polymorph II posseses 8. These differences in interactions between molecules of different polymorphs of ritonavir helps to explain the difference in solubility of the two crystalline forms (40).
The deviations from quality that have occurred in medicines due to polymorphism have accelerated the adoption of new standards on the topic in the phamaceutical industry. They discuss the identification of polymorphs and issues related to the impact and control of polymorphism (41).
But it is worth mentioning that most rules and other official documents that were prepared restrict the discussion to solid forms (often only the solid oral forms) and suspensions. For not contemplating the various dosage forms, these documents leave important gaps. Gaps which if eliminated in the future may decrease the likelihood of further cases such as ritonavir (42-43).
The International Conference on Harmonization issued documents that look to improve the control of polymorphism in new and existing drugs. Among them is the guide known as “Guidance on Specifications: Test Procedures and Acceptance Criteria for New Drug Substances and New Drug Products: Chemical Substances.” (44) discusses various issues, among them polymorphism. Among the decision trees used in this document to indicate the procedures to be followed during the investigation, Tree No. 4 deals with the theme of polymorphism: Researching the need for establishing criteria of acceptance for polymorphism in drugs and medicines (42, 43).
For the Food and Drug Administration (FDA) (45) agency, issues related to polymorphism must be addressed both in the clinical phase (Investigational New Drug - IND) and the stage of registration (New Drug Application - NDA) of innovative medicines (22, 43).
The concern with polymorphism also exists for generic drugs, so that the guide for the industry "Abbreviated New Drug Applications (ANDAs): Pharmaceutical Solid Polymorphism (Chemistry, Manufacturing and Controls Information) – CDER/FDA July 2007” deals just with this theme. The document contains a decision tree subdivided into three smaller trees, which discusses the investigation of polymorphism and the definition of a specification for the API and the drug (22).
The National Health Surveillance Agency (ANVISA) of Brazil has documentation also where the issue of polymorphism is addressed, following international trends, according to Table 1.

A new polymorphic form can be considered an invention, and if it has a significant industrial applicability can be patented, as has occurred with ranitidine, ritonavir, ampicillin, chloramphenicol, celecoxib, novobiocin, griseofulvin, indomethacin, etc. The solid forms that show additional benefits, in terms of physical-technical and physical-chemical properties, and provide enhancement in bioavailability, can be understood as a new drug. Research involving the discovery of new crystalline forms of drugs is important for the patent process, helping innovative companies to maintain the intellectual property of a substance (22, 43).
There are numerous examples where innovative companies acquired patents on a particular polymorphic form, which extended beyond the expiration of the patent for the basic molecule. To their detriment, there is great concern in the pharmaceutical industry of exhausting all the possibilities of polymorphs of a given substance and as such patenting all possible polymorphs as a way of protecting their product. It is known that in current patent laws a new drug is considered newly patented when it exhibits a new polymorphic form, or is co-crystallized with another substance. For the pharmaceutical patent holder of an active principle, this could mean a huge disadvantage if the new polymorph exhibits bioavailability equivalent to an original polymorph of your patent. Therefore, this opens a new dimension to researchers in the pharmaceutical field: the search for an already known polymorph or co-crystal having pharmaceutical bioequivalence or even promoting the pharmacokinetic characteristics with respect to known polymorphs (22, 43).
SELECTION OF THE MOST ADEQUATE POLYMORPHIC FORM FOR THE PRODUCTION OF MEDICINES
Prior determination of the possible crystalline phases of drug candidates comprises a step of great importance for research and drug development of medicines (5, 50), for its successful design requires a broad knowledge of the solid state properties of all materials involved in their formulation including excipients. Although this research does not guarantee that all polymorphs of a substance are discovered, it allows for the verification of potential crystalline phases and ensures that the ideal stage is carefully selected to minimize the risk of late onset of unwanted polymorphs (5).
The choice of an active component in a single polymorphic form for pharmaceutical application must be performed by existing ways of characterizing and evaluating a number of factors such as: hygroscopicity, sensitivity to heat, pressure and shearing, capacity for processing, chemical and physical stability and bioavailability (5), especially the last two as decisive criteria (5, 17). Only with these data can a rational selection of the final solid form be performed (51). However, in the case of polymorphic forms with similar physical-chemical properties, it should be noted that this selection is less important (3, 17). Enalapril maleate, for example, has two polymorphic forms with equivalent solubility and dissolution characteristics, with no effect in terms of bioavailability or the use of one or the other phase in the pharmaceutical formulation (3). This also applies to drugs, such as mefenamic acid, which has polymorphs with different solubilities, but whose difference in absorption and plasma concentration is not significant (18).
Another very important aspect involves the selection of anhydrous or solvated forms, having in mind that during the developmental process of pharmaceutical forms the drugs are frequently exposed to organic and / or aqueous solvents which may lead to the formation of hydrates and solvates (8). These forms usually have different pharmaceutical properties such as bioavailability, stability (8, 10) and mechanical properties (8), which can influence the formulation, the production process and the stability of pharmaceutical products under various storage conditions (10). Therefore, the choice of these forms for pharmaceutical development must be related to the properties mentioned above (8).
The following will discuss the main criteria taken into consideration in the choice of the most suitable polymorphic form for pharmaceutical development.
Stable Form vs. Metastable and Amorphous Forms
Stable Form
Thermodynamically stable polymorphs, as a rule, are preferred for the development of drugs due to their greater crystalline packing density, optimized orientation of the molecules, couplings whether hydrogen or not, crystalline lattice (8), lower internal energy, and consequently greater chemical and physical stability in relation to the metastable forms. Moreover, amorphous forms in general are chemically unstable, have a greater hygroscopicity and tend to stabilize in a crystalline solid (4, 17) due to the lack of a three-dimensional lattice, greater free volume and greater molecular mobility (52).
Chemical stability is an extremely important criterion for drug development, considering that drugs degrade chemically more easily and can lead to a reduction of efficacy or generate toxic metabolites. Presently, physical stability is frequently associated with lower risk of phase conversion, whose most common transformations include amorphization, interconversion, dehydration-hydration, and polymorphic desolvation and transformation which may occur due to variations in humidity, pressure and temperature during production and / or storage of the product (5). Since these conversions can generate polymorphs with different properties and affect chemical stability, mechanical properties and the solubility of these drugs compromising the oral absorption of the drug, the use of the most stable polymorphic forms in order to ensure reproducibility of the efficacy of the product during its shelf life under a variety of storage conditions is preferable (52). This use is particularly important in the production process of suspensions in order that processing may generate polymorphic crystals of different sizes, affecting the stability of the final formulation (18)
However, even with the greater chemical and physical stability of the polymorph of lower energy, adequate control must be exercised during the production process to avoid a possible conversion phase to other polymorphic forms (52).
Metastable and Amorphous Forms
Although a stable polymorphic form is preferably selected for the development of drugs for the reasons cited in the previous section, there are situations that require the use of a metastable crystalline or amorphous form (Table 1) (4, 52). Among these conditions, the most common is to increase the dissolution rate of a medicament due to the low aqueous solubility of the polymorph of lowest energy (52). This is because the metastable and amorphous forms have a higher aqueous solubility, the largest distinction observed between amorphous and crystalline. While the difference in solubility between the crystal forms is typically less than about ten times the difference between amorphous and crystalline materials it can even be hundreds of times (53). For this reason, some drugs are preferably produced with the drug in its metastable form, as in the case of chloramphenicol (53) torsemide (3), phenylbutazone (18) mebendazole (34) and norfloxacin (19) or in amorphous form, in the case of novobiocin (17), cefuroxime axetil (3), tetracycline, and cefalexin (18). It is noteworthy that this choice is also extremely useful for new pharmaceutical molecules in development, bearing in mind that most of them have low water solubility (54). Furthermore, it is also an option for promoting a higher rate of dissolution of a drug in order to bring rapid relief to acute symptoms (52).
Moreover, the selection of an amorphous or metastable form due to the low aqueous solubility of a polymorph of less energy must be evaluated according to the Biopharmaceutical Classification System (BCS). Under this system, only drugs with a rate and a limited extent of absorption by its solubility has a bioavailability and a bioequivalence more likely to be affected by polymorphs of different solubilities. However, it has been verified that there is a lower susceptibility when the rate and extent of absorption is limited by intestinal permeability. Furthermore, differences in solubility of polymorphs hardly affects the bioavailability and bioequivalence of drugs with quick dissolution rates in relation to gastric emptying and when the polymorphic forms are sufficiently high. For this reason drugs which exhibit a high aqueous solubility do not require the establishment of an ideal polymorphic form for increased bioavailability, for example, the situation of ranitidine hydrochloride, terazosin hydrochloride (3) and metropolol (35).
In addition to the increased dissolution rate, other selection criteria are related to the instability of the polymorph of lower energy due to the juxtaposition of reactive groups on adjacent molecules in the crystal lattice, the ease of production and for business purposes, more particularly, patents (52 , 55). As regards the ease of production, it is preferable to use suitable excipients and processing for improving the mechanical properties of the more stable polymorphs instead of choosing these less stable forms (52). This approach will be discussed further in a specific topic.
Despite lower chemical stability of metastable and amorphous forms, their development as a product is possible through a well- planned design, using appropriate excipients and production processes (8, 52). For proper planning thorough knowledge of the characteristics and behavior of the polymorph, proper design of the product and a detailed methodology of control (4) is necessary ensuring the absence of significant risk to the patient, because the conversion phase during shelf-life time may result in an increase of the pharmacokinetic variability and a decrease of the dissolution rate, bioavailability and performance in vivo (52). Ritonavir is one of the best known cases. It was withdrawn from the market in 1998 for having presented changes in dissolution in batches of Novir® capsules, resulting from the appearance of polymorphic Form II after two years of marketing the product in its polymorph Form I which is thermodynamically less stable but with a higher solubility than that of Form II (18, 40).
Solvates versus Hydrates
Solvates
Use of solvates is not recommended for the development of medicines because of the risk of desolvation and release of organic vapors such as impurities or toxic elements of the formulation (5, 14). However, certain substances have a great tendency to form solvates (5) such as spironolactone, which exhibits six polymorphs, with two and four anhydrous solvates (35), besides the classic example of antibacterial sulfathiazole, which has more than 100 solvates described in the literature (5). On the market there are also available medicines produced with some drugs in the form of solvates, namely: darunavir ethanoate (Prezista®), indinavir sulphate ethanoate (Crixivan®), isopropanol sodium warfarin (Coumadin®) (5) and doxycycline hyclate (6).
Accordingly, for the development of safer drugs it is necessary to use Class III solvents such as ethanol, 2-propanol and acetone by virtue of having low potential for toxicity (5, 6). The maximum levels permitted for these solvents in drugs are established in the Guidelines for residual solvents from the International Conference on Harmonization of Technical Requirements for Registration of Pharmaceuticals for Human Use. (56). Another aspect of the formulation that also must be considered is the interaction of molecules of the solvents with the excipients and APIs (8, 52) in order to prevent possible change in its stability and performance (8).
Moreover, solvates can exhibit different physical-chemical and mechanical properties in relation to their corresponding nonsolvated forms and can have positive or negative impact on the pharmaceutical formulation. The solvates of beclomethasone, for example, are quite efficient for micronisation and the formulation of aerosol suspensions and nasal sprays (57).. With regard to in vivo absorption, each form may present a different behavior (27). While monoethanol solvate of prednisolone presents a higher absorption rate than its anhydrous phase, the solvate hemiacetone has practically the same rate (27). Presently, the solvates gliblenclamide obtained with pentanol and toluene present a higher solubility and dissolution rate than their non-solvated polymorphic forms (52). Additionally, solvates present low solubility in their solvents of crystallization (53, 57) due to having a high affinity with the solvent that was incorporated into the crystalline lattice (21) and spontaneous crystallization of solvates produce crystals of lower free energy than their corresponding non-solvated forms (57).
Hydrates
The formation of hydrates is a common phenomenon that occurs with organic molecules due to the ever present nature of water in the environment (5). It is estimated that approximately one third of pharmacologically active substances are capable of forming hydrates (10, 20). This is because the small size of the water molecule favors its incorporation into the voids of the crystalline lattice, and because the multidirectional nature of the hydrogen bonding is also ideal for stabilizing the crystal structures of most molecules (20).
Because many pharmaceutical solids come in contact with water during some stages in the production process, such as crystallization, lyophilization, wet granulation, coating (aqueous film-coating) and spray-drying and during storage (58) it becomes necessary to know about the conditions of stability, particularly humidity and temperature, in which the phases are stable (5). So, similar to polymorphs in general, physical stability and anhydrous forms of hydrates may depend on the relative humidity and / or ambient temperature (5, 8). In addition, some pharmaceutical salts, like sodium and hydrochloride form hydrates more frequently than the "non-salts" due to the propensity of water to bind to ionic sites. On the other hand, the "non-salts" are more likely to form solvates and polymorphs (5).
The selection of hydrates for the development of drugs is complex and must consider a number of factors, namely: solubility, dissolution profile, ability to be processed, dehydration-rehydration behavior and solid state stability (5). Among these factors, it should be noted that hydrated crystals tend to exhibit a lower dissolution profile of the non-hydrated forms (5), since being based on the thermodynamic solubility theory, a solvate is less soluble than when the same compound in its original form is in its crystallization solvent. Thus, hydrates are less water soluble than their corresponding anhydrous form. However, if the formation of the solvate solvent is miscible with water, the solvate will be more water soluble than its anhydrous form. For example, caffeine hydrate has much less water solubility than the solubility of its anhydrous form, however, the hydrate has alcohol solubility much larger than the anhydrous (53).
Despite the lower profile of dissolution, solid form hydrates are preferred for the development of some drugs, such as Cefadroxil (a monohydrate), Paroxetine Hydrochloride (a hemihydrate) (6), Alendronate Sodium, Amoxicillin, Atorvastatin and Pantoprazole (5).
FORMS OF POLYMORPHISM ASSESSMENT APPLIED TO THE PHARMACEUTICAL INDUSTRY
Principle Obtainment Techniques
The process of synthesis of drugs has some critical aspects which favor crystalline transition, such as crystallization, drying, stocking and micronization (5). Although this constitutes one of these aspects, crystallization is a widely used process in the pharma-chemical industry in the purification and separation of compounds, and it is this process that defines the degree of chemical purity and physical properties of the substance which is being isolated, such as size and particle shape, crystalline structure and degree of crystal imperfections of the same (59).
This process is carried out after obtaining a supersaturated solution of the compound and subsequent variation of some parameters such as: sample concentration from the slow evaporation of solvent, temperature, pH and ionic strength. Thus, certain variations can provide thermodynamically favorable environments for the formation of another crystalline structure, aiming to return it to its state of equilibrium (60).
According to Purohit & Venugopalan (2009) (19), obtaining crystals begins with the nucleation phase, which consists in forming the first ordered aggregates, followed by the growth phase. The nucleation step is quite critical, since this is essential for the formation of a metastable or a stable solid form. In supersaturated solutions, for example, any point of nucleation is liable to lead to the formation of crystalline material, but not always to the most stable polymorph (61).
In addition to the rate of nucleation, another critical point in controlling this process is the rate of crystal growth. In this context one should take into account the temperature and the importance of the role of each solvent and impurity present in the process of crystallization, as these have a direct influence on the formation of crystalline structure. Depending on the solvent system being used, the difference in thermodynamic stability of a compound compared to others permits these to be separated, where the more soluble and less stable form is crystallized before (62).
According to Wang et al. (2005) (63) and Cunha (2008) (60), a device often utilized by the pharmaceutical industry is that of an anti-solvent technique known as crystallization by addition of an anti-solvent, which looks to increase the growth rate of a drug with slow growth in organic solvents. The method consists of adding a solvent, which does not solubilize the drug in question (for example water for drugs of low water solubility), to the drug solution in the organic liquid. Thus, Wang et al. (2004) (64) used polyethylene glycol (PEG-300) solvent as an alternative solvent or anti-solvent to replace volatile organic solvents commonly used for performing the crystallization and obtaining of polymorphs. According to the authors, under the conditions of the study, PEG showed the same ability for solubilization as the the 2-propanol and methanol solvents for ibuprofen, salicylic acid and amino acids compounds, evaluating the crystallization of substances as to the induction time of nucleation and the rate of crystal growth. Thus, they observed that the precipitation of salicylic acid and glutamic acid by anti-solvent from the aqueous solution PEG300 or methanol by addition of water indicated a greater induction time for crystallization of PEG solutions, primarily due to higher energy interfacial crystallization. Although the rates of crystal growth were also lower in PEG solutions than in alcohol solutions as well as for asalicylic acid and for glutamic acid, the rates are still quite reasonable for other pratical operations. Still one can observe the same crystal structures of salicylic acid and glutamic acid crystallized as aliphatic alcohols and PEG.
Moreover, Chen and Nan (2011) (65) used surfactants mixed with azanium hexadecyl bromide (trimethyl) and sodium dodecyl sulfate as an antisolvent to mediate the nucleation and crystal growth of CaCO3. Different polymorphic forms were obtained (vaterite and aragonite). Since the adsorption of the surfactant on the surface of the sample is different to the other agents due to the effect of repulsive electrostatic interactions and hydrophobics, the surface energy of the sample was modified by differenet adsorptions of these surfactants. This change induced the difference in metamorphosis and morphology of the samples.
Another classic method consists of cooling a molten sample below its melting point in order that transition can occur between the polymorphic forms upon cooling, since the molecules can arrange themselves differently forming different crystal lattices (66).
The attainment of some suppositories and semi-solid dosage forms, both topical and oral matrix systems are examples of crystallization by cooling (67). In addition, there are numerous specific methods for obtaining polymorphs. Although many of them are based on the principle of crystalization, its specificity occurs by the joining of this technique with new more efficient tools. These methods reduce the activation barrier for nucleation and thus accelerate the crystallization process; particularly, it is useful for substances which are difficult to crystallize (5, 61, 62, 68).
In this context, Capes and Cameron 2007 (69) investigated the effect of the conditions of slow evaporation of a solvent upon the polymorphic crystallisation of paracetamol and observed the obtainment of the metastable crystalline form of the drug. Since the metastable polymorphs present high internal energy and, consequently, a higher rate of dissolution and bioavailability than the stable ones, the use of metastable polymorphs are often required as a strategy to increase of the solubility of slightly hydrosoluble medicines. Although there is the potential risk of polymorphic conversion to the thermodynamically more stable or for other possible polymorphic forms (59) other techniques may be used to verify the stabilization of the crystalline structure, preventing transition as shown by a Capes & Cameron study 2007 (69 ), in which stability was reached from crystallization of the contact surface.
Using the technique of supercritical fluid has been applied to the manufacture of pure polymorphs. This technique is usually performed in a one-step process, where it is possible to isolate different polymorphs by controlling the temperature, pressure, flow and solvent (68). Using high pressure conditions, Tozuka (2003) (70) obtained solid-solid physical transformation of crystals of deoxycholic acid (DCA) using supercritical carbon dioxide.
Despite the different existing crystallization techniques, there is not a method that can provide absolutely reliable isolation of a certain polymorphic form. Therefore, there are a number of efficient methods that assist in the elucidation of the polymorph obtained (61), as discussed below.
Principle Techniques for Identification and Characterization
The considerable variation in properties among different polymorphs, makes the characterization of all polymorphic forms essential to the complete understanding of the structural level of any molecular material (3). Thus, polymorphs can be characterized and differentiated by their physical-chemical properties employing analytical techniques such as: X-ray diffraction, thermal analysis, microscopy and spectroscopy (44).
X-ray diffraction (XRD)
XRD is the most widely used technique in microstructural characterization of crystalline materials. It investigates and distinguishes between different crystalline forms of substances (71)
When the X-ray beam falls on the material, it is elastically spread without loss of energy by the atoms' electrons; if the atoms are arranged in a systematic manner as a crystalline structure, the scattering becomes periodic, forming a characteristic diffraction pattern determined by the crystalline structure of the solid, if the material is amorphous, it has no diffraction pattern, therefore, it is a rapid and efficient method for assessing whether a sample is in an amorphous or crystalline state (72)
It also permits the identification of crystalline phases present in a sample, where it is necessary to obtain the monocrystal to assay the X-ray diffraction, and sometimes it is the only means to determine, among the possible polymorphs of a substance, the structural and the predominant shape, as well as the intermolecular interactions (73, 74).
However, since only one polymotph is thermodynamically stable under a given set of environmental conditions, it is often impossible to obtain crystals of the appropriate size and quality for the analysis of the simple structure of a single crystal. In this case, one must resort to XRD powder where the overlap of the reflection limits information.
However because of the high cost of equipment, the use of the technique becomes impractical for many research laboratories and industries (67).
The X-ray crystallography is, of course, the technique of choice for the direct and definitive identification of different polymorphs. However, it is not always applicable in the case of coexistence of different polymorphs in the same crystal (75).
Thus, the characterization of polymorphs is done generally using other analytical techniques that together generate data capable of confirming the presence of different crystalline forms.
Infrared spectrometry (IR)
Analyses by infrared spectrophotometry are essential for the characterization of polymorphic structures, as its chemical structure indicates whether there is change, or not, in spectral absorption bands of the main functional groups after the crystallization process (76).
Girlando et al. (2010) (75) identified two polymorphs, the R and β-TPB and noticed clear differences between the IR (infrared) spectra of the two phases, confirming that the molecular shape is different.
Lohani et. al. in 2011 (77) used spectroscopic techniques to verify the molecular aggregation of the polymorphic form I of indomethacin in solutions of acetonitrile and ethanol. Their data show clearly the formation of intermolecular interactions (hydrogen bonds) between residual water of the solute and the solvent. They realized that the effects of interactions are important in determining the onset of nucleation in supersaturated solutions that is a key determinant of the relative rate of nucleation and crystal growth of polymorphs, and thus could control the preferential crystallization of polymorphic form I.
Raman scattering spectroscopy
Raman spectroscopy allows for an easier characterization of the structural differences of the polymorph, due to the ability to obtain spectra in the lower vibrational frequencies; it can give vibrational information about the crystal network (78).
The technique provides chemical information similar to IR but complementary since the IR absorption depends on the variation of the molecular dipole moment and the Raman polarizability variation, however, when applied to identification of polymorphs, the technique has several advantages. It is quite suitable for in situ studies that can be performed even in the presence of water or through glass, eliminating the need for a sample preparation (79).
Due to its ease and the current demands of legislation for the control of polymorphic forms, searching for the technique application has been growing and expanding (80).
Raman spectroscopy is widely used in tests for quality control of medicines. The method is fast and perfectly suited to meet the regulatory requirements for monitoring the crystal forms during processing and storage and often able to detect the crystal form present in medicines even when the excipients used are not known (81).
Nuclear Magnetic Resonance of Hydrogen (NMR’H)
The use of NMR in the investigation of polymorphism is easily understood, because if a compound has two polymorphs, a and b, their crystalline forms are conformationally different.. This means that a particular carbon present in form a may have a slightly different molecular geometry when compared to the same carbon in form b, which may give rise to local distinct environments despite having the same atoms bonded together. The difference in the local environment can cause different chemical displacement interactions to the same carbon atom, in two different polymorphic forms (82).
Thus, the NMR parameters can be correlated with structural features such as local subunits, bond distances and angles, or geometries. Moreover, the intensities of the NMR signals are proportional to the number of atoms per unit cell, and the relative occupation of atomic sites can be determined, as well as identifying the amount of existing polymorphs (83).
A major advantage of NMR is the possibility of using this technique in the finished product, i.e. the formulated drug since the chemical displacement of the polymorph is sensitive to the chemical environment and the molecular conformation which allows for the investigation of the conversion of various polymorphs during processing (82).
Shaibat et al. in 2007 (84) discussed the possibility of distinguishing polymorphs of paramagnetic Cu (II) by means of NMR'H and C13 and demontrated that spectra for complex paramagnetic polymorphs have unusually large differences in line widths and spectral positions.
Scanning electron microscopy (SEM)
This technique is very useful for the external structural study of the sample; in the case of polymorphs, it is important for the characterization of surface effects (59).
Sheikhzadeh et al. (2006) (85) characterized form 1 and 2 of buspirone hydrochloride, an anxiolytic drug. The techniques used for characterization included microscopy, thermal analysis, infrared, X-ray diffraction and Raman spectroscopy. Morphologically, form 1 and 2 consist of a plate and columnar crystals, respectively.
In mid-1998, several batches of capsules of ritonavir had differences in the dissolution study and were examined using microscopy and X-ray diffraction; a new polymorph was identified, that´s why the solubility was reduced compared to the original crystal form (40).
Through photomicrographs obtained by SEM to characterize or identify different polymorphic forms, one can observe significant differences in the morphology of the structures.
Thermal analysis
Thermal analysis is the term used to describe analytical techniques that measure the physical-chemical properties of the samples according to temperature or time when the sample is subjected to a controlled temperature variation. Among the various thermoanalytical techniques Thermogravimetry (TG) and Differential Scanning Calorimetry (DSC) are the most used in the pharmaceutical field in the characterization of thermal features of drugs, the determination of purity by compatibility studies of pharmaceutical formulations, the identification of polymorphs, and the evaluation of stability studies of thermal decomposition of drugs and medicines (86, 87).
As to the evaluation of the presence of polymorphisms in drugs by DSC, the result is the emergence of multiple melting peaks related to each crystalline form, the heating rate is critical at this point. Very rapid heating can hide an endothermic curve, while a very slow heating can allow transition or decomposition, making it difficult to visualize the thermal events. Therefore, the search for crystals must be performed initially by means of a study in two heating ratios 2:20° C / min. (16; 88).
Other important aspects for the observation of polymorphs in the DSC curve are: crystalline transition which occurs prior to the melting point of the polymorph of higher melting point, in which the peak will be endothermic for the enantiotropic polymorphs (reversible process) and exothermic for monotropic polymorphs (irreversible), and no weight loss is observed in the TG curve, and the recrystallization process, represented by an exothermic peak which occurs after the melting of the form of a lower melting point (89, 90).
Oliveira and colleagues (2010) (91) promoted the crystallization of simvastatin in different solvents and verified the crystallization of the drug in two forms acicular and tabular. With the aim to identify the polymorphic forms, DSC curves were obtained for the acicular and tabular forms, and there was no duplication of melting points and no appearance of crystalline transition events, indicating no polymorphism. Still, the experimental diffractograms were subjected to adjustment as per the Rietveld method and the structure anticipated for both difractograms is for a tabular crystal for simvastatin. Given this, simvastatin does not present the classic polymorphism, but shows a polymorphism of a morphological type, where the drug has the same unit cell but with different crystal habits due to preferential solute-solvent interactions in the process of crystal growth. This morphological polymorphism does not change the physical-chemical properties and thus, both crystal forms, acicular or tabular form, exhibit the same stability, solubility and reactivity.
Given this fact, this type of polymorphism where crystals with different morphologies are obtained but with the same unit cell, may be in fact due to influence in the growth axis of the nucleus, which is dependent on concentration, temperature and type of solvents used. They can only be identified by scanning electronic microscopy, in DSC curves where they are not identified in multiple melting peaks, while the analysis by XRD is not possible to observe crystallographic differences between the crystalline forms. However, in the XRD differences in intensitiy were observed in specific reflexive planes, suggesting the same unit cell crystals with preferred orientation for the macroscopical development of crystallites in different ways (16).
Silva et al. (2009) (92) used the DSC-photovisual technique to identify and characterize the quality of indinavir sulfate in commercial samples according to their thermoanalytical properties, a method that allows the monitoring of changes produced by a sample temperature range through observation with polarized light microscopy. This method is very useful and is complementary to the DSC for information of compatibility of formulations and identification of polymorphism. Therefore, the photovisual DSC showed the formation of two kinds of solvates with changes in peak melting point and in the melting point of indinavir sulfate. The presence of different amounts of water in the indinavir molecule can alter the thermal and physical-chemical properties (e.g., solubility and bioavailability) of the drug.
Dissolution
The presence of different polymorphs in a formulation can compromise the dissolution of a drug from the pharmaceutical form since different polymorphs often exhibit different solubilities. In some cases the difference is not significant. But when the opposite occurs, the medicines may become less active, inactive or toxic when there is a direct relationship between this parameter and the pharmacological activity. The effect of polymorphism in bioavailability is the most important consequence for the drugs, if bioavailability is measured by dissolution. (34; 67)
The dissolution test determines the mass of the dissolved drug over time, while intrinsic dissolution determines the speed of the drug dissolved. Therefore, when evaluating the different polymorphs in these tests, they usually become more efficient than the solubility in predicting the performance of the different polymorphs in a formulation. An early example presented in the literature, which showed significant changes in bioavailability due to polymorphism was chloramphenicol palmitate. Form B has a therapeutic activity eight times greater than form A (80). This incident triggered discussions about the possibility of occurrence of quality deviations due to polymorphism in various formulations.
Froehlich et al. (34) used infrared spectroscopy and an in vitro dissolution profile to characterize raw materials and tablets (reference and generic) in the Brazilian market. The results obtained show that the three polymorphic forms of mebendazole are present in medicines and two of them in raw materials, suggesting that greater control should be used for the selection of raw materials that exhibit polymorphism, ensuring, through simple and rapid tests, the quality of generic medicines.
Examples like these motivate regulatory authorities of various countries, mainly North America and Europe to deliver resolutions to improve control of polymorphism and decrease the possibility of pursuing changes in the solid form of the drug (59).

INTERFERENCE OF POLYMORPHISM IN THE INDUSTRIAL PROCESSING OF MEDICINES
According to Szterner et al. (2010) (94), newly developed or already established drugs may exhibit unforeseen changes in terms of aqueous solubility and stability of the components of the medicine which has a direct impact on their bioavailability, and, therefore, becoming susceptible, compromising their efficacy and safety.
Since variations in crystal habit of the particles can cause considerable variations in their physical-chemical properties, these are one of the biggest problems of the Pharmaceutical Industry. In this context, to know all possible crystalline forms of the material becomes essential for the rational planning of the development of the pharmaceutical form, thus allowing the design of an efficient manufacturing process for the crystalline form employed (95), aiming to foresee the possible production problems and biological effect, and generating devices to circumvent them.
Therefore, to determine the crystal structure of an active compound by the methods of characterization is an early stage of pharmaceutical development, since this may indicate how easily it can be formulated (50). Variations in the crystalline structure of powders of different polymorphs may cause changes in the technological properties such as plasticity, size and morphology of the particle and hence flow properties, which directly affect the manufacturing process of the product (95, 52).
Caracterization: delineation of the production process
According to Nery et al. (2008) (96), spherical particles present compression and predictable flow by relatively simple models. However, deviations from spherical shape, or form, difficult to be described by simple geometrical figures, possess productive performance more difficult to be predicted. Furthermore, the more stable crystalline structure of a particular molecule exhibits strong intermolecular forces, thus increasing its packing density. Such a characteristic affects the reduction of its ability for deformation, so that after the compression process, tablets are obtained with higher degree of porosity compared to the metastable forms, since these have a lower intensity of intermolecular forces and, consequently, more ease of compression (97).
For this reason, amorphous solids show improved mechanical properties compared to highly crystalline solids, since they consist of disorganized structures and therefore have very weak intermolecular forces, providing them with a great capability for compression. Additionally, they possess other favorable attributes in the pharmaceutical field, since they have a high water solubility and dissolution rate (20).
However, some formulation complications and biological behavior can be observed due to molecular disorganization surrounding these particles once they are thermodynamically unstable and therefore prone to structural change, with tendencies for stability (98). Since small changes in the particle packing density cause significant changes in crystal structure and hence solubility and stability, the use of amorphous particles becomes questionable (99).
Thus, stable crystalline forms tend to be preferred to metastable and amorphous forms (53, 18). However, the stability of these structures usually confers inadequate mechanical properties, as well as limitations in terms of bioavailability. In this context, intervention of pharmaceutical technology is imperative, aiming at the choice of excipients and suitable processes for the crystal form in question.
However, a metastable crystalline form may be sufficiently stable under environmental conditions, making it therefore an excellent alternative to increase aqueous solubility and adjust its physical properties to the processes involved. This phenomenon can be illustrated with graphite and diamond, although consisting of the same molecular structure, the stable form (diamond) requires an activation of high energy and, therefore, graphite (metastable form) is most commonly found in nature (59). Therefore, crystallization techniques are often employed to induce obtaining specific morphology of crystals, in order to optimize the production process and circumvent potential problems of bioavailability. According to Shekunov & York (2000) (100) this device was used in the development of several medicines from drugs such as nitrofurantoin, ibuprofen, octotiamine and acetaminophen.
Moreover, in situations where the metastable and amorphous forms are required and these do not have sufficient stability, it should also make use of pharmaceutical technology aiming, however, at the stabilization of the drug in order to avoid possible conversion of the phase throughout the production process and the medicine storage period (101).
Therefore, the following points of the production process of drugs are discussed that can provide changes to the crystalline form employed, both the active pharmaceutical ingredient and the excipients and can add negative values of the product with respect to their efficacy and safety. Moreover, points will also be discussed that can lead to unintended changes that bring benefits to the process and / or the finished product.
R , D & I and the manufacturing process of medicines: interference of polymorphism and strategies
The industrial steps for obtaining drugs can induce crystalline transition of the different components of the formulation. These transitions chiefly occur due to variations in environmental conditions (such as temperature and pressure) exposing the material to stress conditions, accelerating the crystalline transition of the particles, since in these conditions a new polymorphic form becomes stable (20, 102). Consequently, the physical-chemical characteristics of the finished product are changed, thus reflecting on the unsatisfactory results of quality control tests.
Moreover, this stress may be intentional as metastable crystalline forms may be useful, as previously discussed (20). Thus, knowledge of all possible polymorphic forms within the real conditions of production and storage of the product allows you to design an efficient manufacturing process for a particular polymorphic form (18), thereby aiming to circumvent the environmental conditions that induce undesired crystalline transition or which permits the enjoyment of the desired and conscientious transition.
In this context, Davis et al. (2004) (103) and Saifee et al. (2009) (8) pointed to wet granulation and the drying process as a critical points in the process of production of medications which change the crystalline structure components. According to the authors, the combination of wetting phase and the formation of granules, in addition to kiln drying is sufficient to induce the conversion of crystalline forms, such as with drug Z, the focus of a study by Wang et al. (2010) (104) (Figure 1).

It can be said that there are numerous reports of drugs which undergo crystalline transition during the process of wet and dry granulation (Table 2).

In addition, Guo et al. (2011) (106) highlight the involvement of drug-excipient interaction in crystalline transition. According to Sanchéz et al. (2007) (18) and Varshney et al. (2009) (107) crystalline transitions induced by excipients and / or active ingredients can affect manufacturability and / or disintegration of the dosage form, and in some cases it is necessary to select an alternative crystalline form to eliminate changes in the stability, variations in the dissolution rate and transitions during the process.
Note, therefore that this phenomenon has been observed in some of the excipients whose use is common in pharmaceutical formulations (Table 3).

RESUMO
Neste trabalho foram empregadas as técnicas de DSC e TG/DTG com o objetivo de determinar o comportamento térmico do LPSF/FZ4 isolado e associado com excipientes (amido, βCD, celulose, lactose, estearato de magnésio, aerosil, lauril sulfato de sódio, polissorbato e PVP) e métodos isotérmicos e não-isotérmicos foram realizados para a investigação dos parâmetros cinéticos de decomposição. Através do DSC e tomando como base a equação de Van’t Hoff foi estabelecida uma pureza de 98% para o protótipo. O estudo cinético encontrou uma ordem de reação 1 com Ea 98,22 kJ mol-1, o método isotérmico através da equação de Arrhenius permitiu o cálculo do tempo de estabilidade do LPSF/FZ4, quantificado em 4 meses. As misturas físicas 1:1 dos excipientes com o LPSF/FZ4 mostraram possíveis interações físicas e químicas do protótipo com PVP, polissorbato e lactose. Por outro lado, os resultados do IV não evidenciaram nenhuma modificação química na estrutura da molécula. Mesmo assim, como a curva TG do LPSF/FZ4 associado com a lactose evidenciou uma redução de mais de 40°C da estabilidade do protótipo, foi realizado a cinética de degradação. Tais resultados confirmaram a presença de interação entre o LPSF/FZ4 com a lactose, através da redução de aproximadamente 30% da Ea da reação de decomposição do protótipo mostrado em ambos os métodos cinéticos. O estudo mostrou a importância do uso de métodos termoanalíticos no estudo de caracterização de novos fármacos para a obtenção de parâmetros tecnológicos, possibilitando o desenvolvimento e controle de qualidade de produtos farmacêuticos.

INTRODUÇÃO
A análise térmica está entre as técnicas mais utilizadas (mais de 30 anos) para analisar substâncias de interesse farmacêutico e resolver e ou identificar problemas na área da tecnologia farmacêutica. Revelam informações importantes sobre as propriedades fisicas dos materiais como: estabilidade, compatibilidade, polimorfismo, cinética de decomposição, transição de fase, pureza etc (REZENDE; SANTORO; MATOS et al., 2008, CIDES et al., 2006, ARAÚJO et al., 2003). Estas técnicas permitem uma aquisição rápida de resultados e exigem relativamente simples condições experimentais, no entanto é aconselhável o uso de outras técnicas analíticas (IV, DRX, RMN, MEV, etc) para auxiliar na interpretação dos resultados termoanalíticos (TITA et al., 2011, SALVIO-NETO; NOVA’K; MATOS., 2009).
O estudo de compatibilidade fármaco excipiente representa uma importante etapa no estágio de préformulação para o desenvolvimento de uma forma farmacêutica (PINTO et al., 2010). As interações físicas e químicas entre os ingredientes ativos e excipientes da formulação podem afetar as características químicas, estabilidade e biodisponibilidade do medicamento e, portanto, seu efeito terapêutico e eficácia. De fato, os métodos térmicos, possibilitam a detecção de interações físicas, bem como a formação de misturas eutéticas ou adsorção entre os compostos ativos e os excipientes, como por exemplo, na curva DSC pode-se observar possíveis interações de acordo com a mudança de aparência, ou o desaparecimento dos picos endotérmicos ou exotérmicos e ou variações nos valores de entalpia correspondente a mistura fármaco excipiente (TITA et al., 2011, BHARATE; BHARATE; BAJAJ., 2010).
Os estudos de cinética de degradação tornaram-se um ponto crucial em relação à análise térmica, onde o objetivo principal é determinar o mecanismo de reação de decomposição e os parâmetros da equação de Arrhenius (energia de ativação, fator de frequencia e ordem de reação). Estes dados podem fornecer informações preciosas sobre as condições de armazenamento, especialmete tempo de vida útil, meia vida e vida de prateleira. O conhecimento de tais parâmetros faz parte das atividades de pesquisa durante o desenvolvimento de um novo produto farmaceutico (SOVIZI., 2010, TITA et al., 2009, TITA et al., 2008, OLIVA; LLABRÉS; FARIÑA et al., 2006).
Trabalhos desenvolvidos pelo Laboratório de Planejamento e Síntese de Fármacos do Departamento de Antibióticos da UFPE, sintetizaram o LPSF/ FZ4, com a finalidade de atuar como agente na terapia contra esquistossomose. O LPSF/FZ4 é um derivado imidazolidínico e sua atividade contra esquistossomose foi realizada por Oliveira et al (2004), mostrando -se o protótipo mais ativo dentre os fármacos do grupo de novas séries de imidazolidinas.

Sendo assim, por se tratar de uma mólecula nova e pela pouca informação disponível na literatura, o objetivo deste estudo foi empregar TG/DTG e DSC para o estudo do comportamento térmico, compatibilidade com excipientes e cinética de decomposição do LPSF/FZ4 sob condições isotérmica e não-isotérmicas, afim de estabelecer padrões de comportamento para esse candidato a fármaco a ser veiculado em uma determinada forma farmacêutica.
EXPERIMENTAL
MATERIAL
LPSF/FZ4 (Lot. 01), 3-(4-cloro-benzil)-5-(4-nitro-benzilideno)-imidazolidina-2,4-diona, foi fornecido pelo Laboratório de Planejamento de Síntese de Fármacos (LPSF) com pureza de 98% estimada por DSC (Calorimetria exploratória diferencial). Os excipientes usados foram Celulose microcristalina (CMC) (Blanver Farmoquímica Ltda®), Beta ciclodextrina (CD) (Roquete ®), Polivinilpirrolidona (PVP) (Xiamem®), Septrap 80 (Polissorbato) (Seppic ®), Dióxido de silício coloidal (Aerosil) (Henkel®), Laurilsulfato de sódio (LSS) (Nuclear®), amido (Colorcon®), estearato do magnésio (Ind. Química Anastácio S/A®), lactose (Pharma Nostra®).
MÉTODOS
O fluxograma (Figura 2) apresenta o protocolo utilizado para a avaliação da estabilidade térmica do LPSF/FZ4. Em primeiro lugar, medidas de calorimetria exploratória diferencial (DSC) e termogravimetria (TG), somada a investigação da cinética de degradação térmica dinâmica e isotérmica foram realizadas para caracterização do protótipo, posteriormente para a análise da interação fármaco excipiente, misturas físicas binárias foram selecionadas para rápida interpretação. A investigação das possíveis interferências responsáveis pelas alterações no perfil térmico do composto foi procedida pela cinética de degradação e analise no Infravermelho.

Calorimetria Exploratória Diferencial (DSC)
As curvas de DSC para o fármaco foram obtidas por meio de um calorímetro da marca Shimadzu®, modelo DSC-60, em atmosfera de nitrogênio em fluxo de 50 mL.min-1, sendo a massa das amostras analisadas em torno de 2,0 mg (± 0.2), acondicionadas em cápsula de alumínio, na razão de 10°C/min, até a temperatura de 600ºC. As determinações foram realizadas em triplicata. Realizou-se a calibração do DTA e do DSC via ponto de fusão do padrão Índio (156,6ºC) e Zinco (419,6ºC). O fluxo de calor e entalpia foram calibrados via ponto de fusão do Índio (28,59 ± 0,3 J.g-1), sob as mesmas condições das amostras.
Termogravimetria (TG)
A caracterização termoanalítica através de TG foi realizada em triplicata por meio de termobalança, modelo TGA Q60 da marca Shimadzu®, em atmosfera de nitrogênio com fluxo de 50 mL.min-1, sendo a massa da amostra de cerca de 3 mg (±0,4) de LPSF/FZ4, acondicionadas em cadinho de alumina na faixa de temperatura de 25-600˚C na razão de 10°C/min. A calibração do instrumento foi verificada antes dos ensaios empregando-se um padrão de oxalato de cálcio monoidratado, conforme norma ASTM E1582-93 (The American Society for Testing and Materials, 1993) (ARAÚJO et al., 2006, STORPITS et al., 2009, KLANCNIK et al., 2010).
Estudo cinético de decomposição
A investigação cinética de degradação não-isotérmica do LPSF/FZ4 foi obtida a partir dos dados de TG pela aplicação do método de Ozawa. Foram utilizadas razão de aquecimento 2,5, 5,0, 7,5, 10 e 12,5°C.min-1, faixa de temperatura entre 30-600°C, em porta amostra de alumina com aproximadamente 2 mg de amostras e atmosfera dinâmica de nitrogênio em (50 mL•min-1). Para o estudo cinético isotérmico, as curvas TG foram obtidas a partir do aquecimento das amostras até as temperaturas de 260, 270, 280, 290 e 300 °C, e mantidas em condições isotérmicas em atmosfera de N2 (50 mL min–1) durante o tempo necessário para uma perda de massa de 10% ± 0,5 em porta amostra de alumina com aproximadamente 6 mg de amostras.
Estudo de compatibilidade
Amostras de LPSF/FZ4 e cada excipiente foram individualmente pesados em quantidades iguais e colocados em recipientes de vidro âmbar para dar peso final de 60 mg. As misturas binárias 1:1 (LPSF/FZ4:excipiente) foram preparada na relação p/p através de uma simples mistura. Foi adicionada 3 mg da mistura binária no porta-amostra de aluminio hermeticamente fechada do DSC e submetidas as mesmas condições já descritas.
Espectrofotometria de absorção na região do infravermelho (IV)
Os espectros de infravermelho foram obtidos utilizando o equipamento PerkinElmer® (Spectrum 400) com dispositivo de reflectância total atenuada (ATR) com cristal de seleneto de zinco. As amostras do protótipo e das misturas fármaco-excipiente analisadas foram transferidas diretamente para o compartimento do dispositivo de ATR, sendo o resultado obtido da média de 10 varreduras, de 4000 a 650 cm-1 na resolução de 4 cm-1.
RESULTADOS E DISCUSSÃO
Caracterização termoanalítica
A curva DSC do LPSF/FZ4 mostrou um pico endotérmico correspondente ao evento do processo de fusão, na faixa de temperatura de 228°C a 234°C com um consumo de energia de -178 Jg-1, seguido por processo de decomposição. O resultado foi confirmado pela análise de TG e pode ser melhor visualizado através da aplicação da primeira derivada (DTG), que o processo de decomposição térmica ocorre em uma fase na faixa de temperatura entre 323°C e 370°C, com perda de massa de 60,85% seguido por carbonização (Figura 3).

Determinação da pureza
Devido o LPSF/FZ4 ser até o momento um protótipo, e não possuir SQR (Substância Química de Referência) ou substâncias similares é importante à determinação da pureza e essa ferramenta é muito importante para os estudos de pré-formulação. A pureza foi determinada através do desvio da linearidade do evento de fusão, o qual ocorre pela presença de impurezas, calculada através da equação de Van’t Hoff (ARAÚJO et al., 2010). Logo, a pureza média do LPSF/FZ4 por DSC mostrou-se em torno de 98,24%.
Na Figura 4 observa-se um gráfico denominado 1/F (T x 1/F), que mostra a curva de temperatura de fusão versus o inverso da fração fundida, normalmente uma reta que permite a correção das áreas parciais. A inclinação da reta é proporcional ao teor de impurezas e a interseção com o eixo das ordenadas corresponde à temperatura do material puro (STORPITS et al., 2009). Neste caso, o LPSF/FZ4 funde em 229°C e apresenta um valor médio do teor de pureza já descrito acima.

Estudo cinético de termodecomposição do LPSF/FZ4
Há duas maneiras de determinar os parâmetros cinéticos, uma utiliza o método convencional de estudo isotérmico por TG que através da equação de Arrhenius determina os parâmetros cinéticos e o estudo não-isotérmico que utiliza o método de Ozawa originado pela aproximação linear com base em cálculo integral da equação de Arrhenius (SALVIO-NETO; MATOS., 2011, SOVIZI., 2010).
Os dados cinéticos não-isotérmicos foram determinados pelo plotagem da perda de massa versus temperatura de cinco curvas TG em diferentes razões de aquecimento (Figura 5). Foi utilizado o método de Ozawa, o mesmo é baseado nos cálculos da integral a partir da equação da Arrhenius para obtenção dos parâmetros cinéticos no início da etapa de decomposição térmica em torno de 340-370° C do LPSF/ FZ4: Ea (energia de ativação) 98,22 kJ/mol, A (fator de frequencia) 5,28 x107 min-1 e ordem de reação 1,0.

O gráfico de Ozawa demonstra uma boa correlação linear entre as cinco razões de aquecimento (FELIX; CIDES DA SILVA; MATOS., 2009). As cinco curvas TG indicam que a perda de massa entre 250°C e 400°C, ocorreu em uma única etapa, o que corrobora com a ordem de reação assumida de primeira ordem, uma vez que uma única reação contribui para a perda de massa, ou seja, há apenas uma reação significativa ocorrendo e não é afetado por processos concorrentes (HOWELL; RAY., 2006). Mostram também que as curvas de TG são deslocadas para maiores temperaturas, quando as razão de aquecimento aumentam. (BERTOL et al., 2010, CIDES et al., 2006)
O método isotérmico usa comumente a mesma razão de aquecimento e as temperaturas são mantidas constantes na região de interesse, ou melhor, antes do processo de decomposição, como mostra a curva TG/DTG (Figura 3) para monitorar a cinética da reação de decomposição térmica no estado sólido, portanto, o tempo de decomposição é estimado para um determinado intervalo de perda de massa (ALVES et al., 2010).
Os dados apresentados na Figura 6 mostram as isotermas utilizadas na determinação da energia de ativação do LPSF/ FZ4. Esta foi realizada aquecendo a amostra a 260, 270, 280, 290 e 300°C e mantidas constantes sob uma atmosfera dinâmica de nitrogênio (50 mL.min-1) por um tempo suficiente para a perda de massa em pelo menos 10%. Estas curvas mostram que quanto maior a temperatura, menor será o tempo necessário para a perda de massa.

Estas curvas foram utilizadas para obter o gráfico de lnt vs a recíproca da temperatura 1/T (K-1) para o LPSF/FZ4 (Figura 7) também chamado de gráfico de Arrhenius, onde a equação da reta foi obtida por regressão linear (y = ax b), para avaliar a validade do modelo cinético, menssurar a linearidade e o coeficinete de correlação (r) (RODRIGUES et al., 2005). O Valor da energia de ativação para o LPSF/FZ4 foi de 174,13 KJ mol–1, calculada a partir do produto do coeficiente angular da inclinação da reta com a constante geral dos grases R (8,314 J mol-1 K-1).
Calculou-se a estabilidade (em dias) com base na equação de Arrehenius, utilizando 25°C como padrão de temperatura ambiente. O resultado foi um tempo estimado de estabilidade térmica de 132 dias, ou seja, aproximadamente quatro meses para um decaimento de 10% de massa.

Estudo de compatibilidade
O comportamento das misturas binárias mostraram pequenas mudanças na forma do pico com poucas variações da temperatura de fusão, indicando ausência de incompatibilidade na maioria dos casos. Os dados TG e DSC obtidos no estudo de compatibilidade estão demonstrados na Tabela 1.

Para a mistura binária LPSF/FZ4:PVP ocorreu o desaparecimento do ponto de fusão do protótipo (Figura 8), isso é indicativo de uma forte interação, em virtude do aquecimento, mas não necessariamente correspondente a incompatibilidade. O comportamento da mistura evidencia a missibilidade do protótipo no PVP fundido ocorrendo total solubilização do LSPF/FZ4, representada por um pico alargado e de baixa intensidade, devido a grande variação dos pontos de fusão individuais.
Na curva DSC da mistura binária de LPSF/FZ4:Polissorbato (Figura 8), houve uma redução considerável da temperatura de fusão de mais de 10°C do início da temperatura de fusão do protótipo, que pode ser indicativo de ocorrência de interação fármaco-excipiente. De acordo com as curvas TG/DTG (Figura 9) o início da temperatura de decomposição diminuiu de 323 para 313°C.

A curva DSC da lactose mostra um pico endotérmico em 145°C correspondente à perda de água, pico exotérmico em 173°C referente a transição cristalina, e um evento endotérmico em 215°C correspondente ao ponto de fusão, e logo seguida a decomposição térmica (CIDES et al., 2006). Na mistura LPSF/FZ4:Lactose, observou-se um leve retardo do início da temperatura de fusão do protótipo, no entanto a curva TG da mistura binária (figura 8) revelou interações entre o protótipo e a lactose, que pode ser de natureza física. Este fato é justificado porque a curva TG (Figura 9) mostrou a antecipação da temperatura de decomposição térmica em torno de 40 ° C abaixo da degradação do prototipo. Tais resultados tambem podem ser observados em outras aminas e amidas como glimepirida, primaquina, glipzida e glibenclamida ( SALVIO-NETO; MATOS., 2011; BERTOL et al., 2010; CIDES et al., 2006).

O espectro na região do IV foi usado como técnica suplementar de ordem investigativa das possíveis interações químicas entre o protótipo e os excipientes e para confirmar os resultados obtidos pela análise térmica.
A Figura 10 mostra os espectros IV do LPSF/FZ4 e da mistura física LPSF/FZ4:Polissorbato, a mesma mostra a presença de bandas características correspondentes ao protótipo e ao excipiente. Não ocorreu aparecimento e desaparecimento de novas bandas, nem alteração na intensidade das bandas no espectro, confirmando nunhuma mudança na estrutura do protótipo ( TITA et al., 2011). Resultado similar tambm pode ser observado para a mistura fisica LPSF/FZ4:Lactose.

As Figuras 11 e 12 mostram as curvas TG da cinética de decomposição sob condições isotérmicas da degradação do LPSF/FZ4 associado com a lactose. As amostras foram mantidas em 200, 205, 210, 215 e 220°C para 10% de perda de massa, representado na Figura 11 a correlação entre a perda de massa e temperatura, bem como, com os dados do experimento e a partir do método de regressão linear, foi construído o gráfico de Arrhenius (Figura 12).

A superposição das curvas TG obtidas em diferentes razões de aquecimento (2,5; 5; 7,5; 10; 12,5°C/min) para a cinético de decomposição sob condições não isotermicas (método de Ozawa) para a degradação do LPSF/FZ4 e lactose são mostradas na Figura 13. A Tabela 2 lista os valores dos parâmetros cinéticos obtidos pelo estudo não-isotérmico e a energia de ativação (Ea) e o coeficiente de correlação (r) obtidos pelo estudo isotérmico.

Foi observado uma redução na Ea obtida pelo método de Ozawa do protótipo associado com a lactose (54,95 kJ mol-1) quando comparado ao protótipo sozinho (98,22 kJ mol-1), da mesma forma também pôde-se observar para o estudo isotérmico (174,13 – 148,34 kJ mol-1), representando aproximadamente uma diferença de 30% na energia de ativação. Esses resultados confirmam a baixa estabilidade do protótipo na presença da lactose corroborando com os resultados obtidos no estudo de compatibilidade por análise térmica e espectroscopia na região do IV (SALVIO-NETO; MATOS., 2011).
CONCLUSÃO
O estudo do comportamento térmico por técnicas termoanalíticas empregadas no presente trabalho permitiu fornecer informações detalhadas sobre uma nova entidade química mesmo em estágios mais iniciais da descoberta quando da identificação até o desenvolvimento de uma formulação. A calorimetria exploratória diferencial permitiu identificar o LPSF/FZ4 através de sua faixa e entalpia de fusão, fornecendo ainda dados quantitativos sobre sua pureza.
A comparação entre os métodos isotérmico e não isotérmico mostrou uma boa concordância para os valores dos parâmetros cinéticos (energia de ativação) demostrando alta estabilidade térmica do protótipo. A decomposição cinética ocorreu em velocidade constante, ordem um (uma única reação que contribui para a perda de massa), ou seja, há apenas uma reação significativa ocorrendo e não é afetado por processos concorrentes.
O resultados do estudo de compatibilidade de algumas misturas binárias foram estudados usando TG / DTG, DSC, IV e cinética de decomposição. Os resultados mostraram a utilidade da análise térmica como um método rápido e conveniente na realização de uma triagem de candidatos a excipiente durante os estudos de preformulação, porque permitiu a demonstração de interações ou incompatibilidade fármaco-excipiente. Nestes estudos, foi observado possível interação do LPSF/FZ4 com a lactose, confirmada através do estudos cinéticos (isotérmico e não-isotérmico) que indicaram uma diminuição de aproximadamente 30% do valor da energia de ativação da degradação do protótipo puro e associado com a lactose, mostrando uma diminuição na estabilidade da fármaco.
Sendo assim, o presente estudo contribuiu para a seleção de excipientes apropriados para o desenvolvimento de uma formulação segura e estável, mostrando através dos resultados da compoatibilidade e das cinéticas que a associação do LPSF/FZ4 com a lactose não é recomendado para o desenvolvimento de uma forma farmacêutica sólida.
Desta forma a utilização da análise térmica traduz uma alternativa de indubitável interesse no campo farmacêutico com o objetivo de prever rapidamente e com baixo custo a estabilidade a longo prazo de um candidato a insumo farmacêutico e acelerar a sua comercialização.

SUMMARY
In this study DSC and TG/DTG techniques were employed with the objective of determining the thermal behavior of isolated LPSF/FZ4 associated with excipients (amide, βCD, cellulose, lactose, stearate of magnesium, aerosil, sodium lauryl sulfate, polysorbate and PVP) and isothermal and non-isothermal methods were carried out for the investigation of kinetic parameters of decomposition. Through DSC and taking the van't Hoff equation as a basis a pureness of 98% was established for the prototype. The study found a kinetic reaction order with 1 Ae 98.22 kJ mol -1,the isothermal method using the Arrhenius equation enabled the calculation of the time of stability of LPSF/FZ4, measured in 4 months. Physical mixtures of excipients with 1:1 LPSF/FZ4 showed possible physical and chemical interactions of the prototype with PVP, lactose and polysorbate. Moreover, the results of the IR did not show any change in the chemical structure of the molecule. Even so, as the TG curve of LPSF/FZ4 associated with the lactose showed a decrease of more than 40°C the stability of the prototype, degradation kinetics were achieved. These results confirmed the presence of interaction between the LPSF/FZ4 with lactose, by reduction of approximately 30% of Ae and that the decomposition reaction of the prototype showed both kinetic methods. The study demonstrated the importance of using thermal analytical methods in characterization studies of new drugs for obtaining technological parameters, enabling the development and quality control of pharmaceutical products.
INTRODUCTION
Thermal analysis is among the most widely used techniques (over 30 years) for analyzing substances of pharmaceutical interest and resolving and/or identifying problems in the field of pharmaceutical technology. They reveal important information about the physical properties of materials such as: stability, compatibility, polymorphism, decomposition kinetics, phase transition, purity, etc. (Rezende; SANTORO; MATOS et al., 2008, CIDES et al., 2006, Araújo et al., 2003). These techniques allow a rapid acquisition of results and require relatively simple experimental conditions, however it is advisable to use other analytical techniques (IR, XRD, NMR, SEM, etc.) to assist in the interpretation of thermal analytical results (TITA et al., 2011 , SALVIO-NETO; NOVA'K; MATOS., 2009).
The drug-excipient compatibility study represents an important step in the preformulation stage for developing a pharmaceutical form (Pinto et al., 2010). The physical and chemical interactions between the active ingredients and excipients in the formulation can affect the chemical characteristics, stability and bio-availability of the drug and thus their therapeutic effect and effectiveness. In fact, thermal methods, enable detection of physical interactions and the formation of eutectic mixtures or adsorption between the active compounds and excipients, for example, in the DSC curve possible interactions can be observed according to the change of appearance or disappearance of endothermic or exothermic peaks and/or variations in the values of the enthalpy corresponding to drug excipient mixture (TITA et al. May 2011, BHARATE; BHARATE; BAJAJ., 2010).
The degradation kinetic studies have become a crucial point in relation to thermal analysis, where the main objective is to determine the mechanism of decomposition reaction and the Arrhenius equation parameters (activation energy, frequency factor and reaction order). These data can provide valuable information about storage conditions, especially useful lifetime, half-life and shelf life. The knowledge of these parameters is part of the research activities during the development of a new pharmaceutical product (SOVIZI., 2010, TITA et al., 2009, TITA et al., 2008, OLIVA; Llabres; Farina et al., 2006).
Work developed by the Laboratory of Drug Synthesis and Planning Department of Antibiotics at FUPE, synthesized LPSF/FZ4, in order to act as an agent in the therapy against schistosomiasis. LPSF/FZ4 is a imidazolidinic derivative and its activity against schistosomiasis was performed by Oliveira et al (2004), showing that the prototype drugs most active among the group of new series of imidazolidines.

Therefore, because it is a new molecule and because of the paucity of information available in the literature, the aim of this study was to use TG/DTG and DSC to study thermal behavior, compatibility with excipients and decomposition kinetics of LPSF/FZ4 under isothermal and non-isothermal conditions, in order to establish standards of conduct for this drug candidate to be transmitted in a particular dosage form.
EXPERIMENTAL
MATERIAL
LPSF/FZ4 (Lot. 01) 3 - (4-chlorobenzyl) -5 - (4-nitro-benzylidene)-imidazolidine-2 ,4-dione, was provided by the Laboratory of Drug Synthesis Planning (LPSF) with 98% purity estimated by DSC (differential scanning calorimetry). The excipients used were micro-crystalline cellulose (MCC) (Blanver Pharmochemical Ltd.®), Beta cyclodextrin (BC) (Roquete ®) Polyvinylpyrrolidone (PVP) (Xiamem® ), Septrap 80 (Polysorbate) (SEPPIC ® ), Colloidal silicon dioxide (Aerosil) (Henkel®), Sodium Lauryl Sulfate (SLS) (Nuclear®) starch (Colorcon ®), Magnesium stearate (Anastácio Chemical Industry®), Lactose (Pharma Nostra ® ).
METHODS
The flowchart (Figure 2) shows the protocol used for evaluating the thermal stability of LPSF/FZ4. First, measurements of differential scanning calorimetry (DSC) and thermogravimetry (TG) coupled with the investigation of the kinetics of isothermal and dynamic thermal degradation were performed to characterize the prototype, later for analysis of drug-excipient interaction, physical binary mixtures were selected for rapid interpretation. The investigation of possible interferences responsible for changes in the thermal profile of the compound was preceded by the degradation kinetics and infrared analysis.

Differential Scanning Calorimetry (DSC)
The DSC curves for the drug were obtained using a Shimadzu® brand calorimeter, Model DSC-60 under an atmosphere of nitrogen flow of 50 ml min-1, the mass of samples around 2.0 mg (± 0.2), packed in aluminum capsules, at the ratio of 10°C/min, up to a temperature of 600°C. The determinations were performed in triplicate. The calibration of DTA and DSC was carried out via the melting point pattern of Indium (156.6°C) and zinc (419.6°C). The heat flux and enthalpy were calibrated via the melting point of Indium (28.59 ± 0.3 J.g-1), under the same conditions as the samples.
Thermogravimetry (TG)
The thermal analytic characterization of TG was performed in triplicate by thermobalance, Shimadzu® TGA Q60 model, in a nitrogen atmosphere with a flow of50 ml/min-1, where the sample mass of about 3 mg (± 0.4) of LPSF/FZ4, packed in aluminum oxide crucible at a temperature range of 25-600˚C at the ratio of 10°C/min. Calibration of the instrument was checked prior to testing by employing a standard calcium oxalate monohydrate according to ASTM E1582-93 (The American Society for Testing and Materials, 1993) (Araujo et al. 2,006, STORPITS et al. 2009, KLANCNIK et al., 2010).
Decomposition Kinetic Study
Non-isothermal degradation kinetics research of LPSF/FZ4 was obtained from TG data by the application of the Ozawa method. Heating rate used was 2.5, 5.0, 7.5, 10 and 12.5°C.min -1, The temperature range of 30-600°C in a aluminum oxide sample port with about 2 mg of samples and a dynamic atmosphere of nitrogen (50 ml/min-1). For the kinetic isothermal TG study, curves were obtained by heating the samples until temperatures of 260, 270, 280, 290 and 300°C, and maintained at isothermal conditions in an atmosphere of N 2 (50 ml min -1) during the necessary time for a mass loss of 10% ± 0.5 in the aluminum oxide sample port with approximately 6 mg samples.
Compatibility Study
Samples of LPSF/FZ4 and each excipient were individually weighed and placed in equal quantities in amber glass containers to give the final weight of 60 mg. The binary mixtures 1:1 (LPSF/FZ4: excipient) were prepared at the ratio p/p by simple mixing. 3 mg of the binary mixture were placed in the aluminum hermetically sealed DSC specimen holder and subjected to the same conditions already described.
Absorption spectroscopy in the infrared (IR) region.
The infrared spectra were obtained using a PerkinElmer® (Spectrum 400) equipment with an attenuated total reflectance (ATR) device with a zinc selenide crystal. The samples of the prototype and drug-excipient mixtures analyzed were directly transferred to the housing of the ATR device and the result obtained by averaging 10 scans, from 4000 to 650cm-1 in resolution of 4 cm -1 .
RESULTS AND DISCUSSION
Thermal analytic characterization
The DSC curve of LPSF/FZ4 showed an endothermic peak corresponding to the event of the melting process in the temperature range from 228°C to 234°C with a power consumption of -178 Jg -1, followed by the decomposition process. This result was confirmed by TG analysis and can best be visualized by applying the first derivative (DTG), in which the process of thermal decomposition occurs in a phase at the temperature range between 323°C and 370°C, with a loss of mass of 60.85% followed by carbonization (Figure 3).

Determination of Purity
Due to LPSF/FZ4 being a prototype, and having no CSR (Chemical Reference Substance) or similar substances it is important in determining purity and this tool is very important for preformulation studies. Purity was determined by the deviation from linearity of the fusion event, which occurs by the presence of impurities, calculated using the van't Hoff equation (Araujo et al., 2010). Therefore, the average purity of LPSF/FZ4 by DSC was found to be about 98.24%.
In Figure 4 a graph called 1/F (T x 1/F) can be seen, which shows the melting temperature curve versus the inverse of the melted fraction, usually a straight line that allows for the correction of partial areas. The slope of the straight line is proportional to the impurity content and the intersection with the vertical axis corresponds to the temperature of the pure material (STORPITS et al., 2009). In this case, LPSF/FZ4 melts at 229°C and has an average value of purity content already described above.

Kinetic study of the thermal decomposition of LPSF/FZ4
There are two ways to determine the kinetic parameters, a conventional method uses isothermal study by TG using the Arrhenius equation that determines the kinetic parameters and the non-isothermal study using the Ozawa method originated by the linear approximation based on integral calculus of the Arrhenius equation (SALVIO-NETO; MATOS., 2011, SOVIZI., 2010).
The non-isothermal kinetics data were determined by plotting the weight loss versus temperature of five TG curves at different ratios of heating (Figure 5). The Ozawa method was used, which is based on integral calculus beginning with the Arrhenius equationfor the acquisition of kinetic parameters in the early stage of thermal decomposition at around 340-370°C of LPSF/FZ4: Ae (activation energy) 98.22 kJ/mol, A (frequency factor) 5.28 x107 min-1 and a reaction order 1.0.

The five TG curves indicate that the weight loss between 250°C and 400°C, took place in a single step, which confirms the assumed reaction order of the first order, since a single reaction contributes to the loss of mass, i.e., there is only one significant reaction occurring and it is not affected by concurrent processes (HOWELL; RAY., 2006). Also they show that TG curves are shifted to higher temperatures when the heating rate increases. (BERTOL et al. 2010, CIDES et al. 2006)
The isothermal method commonly uses the same heating rate and the temperatures are kept constant in the region of interest, or rather, before the decomposition process, as shown in the TG/DTG curve (Figure 3) to monitor the kinetics of the thermal decomposition reaction in the solid state, therefore, the decomposition time is estimated for a given range of loss of mass(Alves et al., 2010).
The data presented in Figure 6 show the isotherms used in determining the activation energy of LPSF/FZ4. This was performed by heating the sample to 260, 270, 280, 290 and 300°C and kept constant under a dynamic atmosphere of nitrogen (50 ml/min-1) for sufficient time for the weight loss of at least 10%. These curves show that the higher the temperature, the less will be the necessary time for the loss of mass.

These curves were used to obtain the lnt graph of the reciprocal temperature versus 1/T (K-1) for LPSF/FZ4 (Figure 7) also called the Arrhenius plot, where the straight line equation was obtained by linear regression (y = ax b) to evaluate the validity of the kinetic model, and measure the linearity and the correlation coefficient (r) (Rodrigues et al. 2005 ). The value of activation energy for LPSF/FZ4 was 174.13 KJ mol -1, calculated from the product of angular coeffiicient of the straight line slope with the general constant of R gases (8.314 J mol-1 K-1).
The stability (days) based on the Arrehenius equation using 25°C as a standard temperature was calculated. The result was an estimated time of thermal stability of 132 days or about four months for a decay of 10% by weight.

Compatibility Study
The behavior of binary mixtures showed little change in the shape of the peak with little variation of melting temperature, meaning no incompatibility in most cases. The data obtained in the TG and DSC compatibility study are shown in Table 1.

For the binary mixture LPSF/FZ4: PVP at the melting point of the prototype vanished (Figure 8), this is indicative of a strong interaction, due to heating but not necessarily corresponding to incompatibility. The behavior of the mixture shows miscibility of the prototype in molten PVP with total solubility of LSPF/FZ4 occurring, represented by a broad peak and low intensity, due to large variation in individual melting points.
In the DSC curve of the binary mixture of LPSF/FZ4: Polysorbate (Figure 8), there was a considerable reduction of the melting temperature of over 10°C from the beginning of the melting temperature of the prototype, which can be indicative of the occurrence of drug-excipient interaction. According to the TG/DTG curves (Figure 9) the onset of the decomposition temperature decreased from 323 to 313°C.

The DSC curve of lactose shows an endothermic peak at 145°C corresponding to loss of water, exothermic peak at 173°C refers to crystalline transition and an endothermic event at 215°C corresponding to the melting point and then immediately thermal decomposition (CIDES et al., 2006). In the mixture LPSF/FZ4: Lactose, there was a slight delay in onset of the melting temperature of the prototype, but the TG curve of the binary mixture (Figure 8 ) showed interactions between the prototype and lactose, which may be physical in nature. This fact is justified because the TG curve (Fig. 9) showed the anticipation of the thermal decomposition temperature of around 40°C below the degradation of the prototype. These results can also be observed in other amines and amides such as glimepiride, primaquine, glipzide and glibenclamide (SALVIO-NETO; MATOS., 2011; BERTOL et al., 2010; CIDES et al., 2006).

The spectrum in the IR region was used as a supplementary investigative technique of possible chemical interactions between the prototype and the excipients and to confirm the results obtained by thermal analysis.
Figure10 shows IR spectra of LPSF/FZ4 and the physical mixture of LPSF/FZ4: Polysorbate, with the presence of characteristic bands corresponding to the prototype and the excipient. The appearance and disappearance of new bands did not occur nor was there a change in intensity of the bands in the spectrum, confirming no change in the structure of the prototype (TITA et al. May 2011). A similar result can also be observed in the physical mixture LPSF/FZ4: Lactose.

Figures 11 and 12 show the TG curves of the decomposition kinetics under isothermal conditions of degradation of LPSF/FZ4 associated with lactose. The samples were maintained at 200, 205, 210, 215 and 220°C at 10% mass loss, represented in Figure 11 is the correlation between weight loss and temperature as well, the Arrhenius plot was constructed with the data from the experiment and linear regression method (Figure 12).

The TG superposition of the curves obtained at different heating ratios (2.5, 5, 7.5, 10, 12.5°C/min) for kinetic decomposition under non-isothermal conditions (Ozawa method) for the degradation of LPSF/FZ4 and lactose are shown in Figure 13. Table 2 lists the values of the kinetic parameters obtained by non-isothermal study and the activation energy (Ae) and the correlation coefficient (r) obtained by isothermal study.

A reduction in the Ae obtained by the Ozawa method of prototype associated with lactose was observed (54.95 kJ mol -1 ) when compared to the prototype alone (98.22 kJ mol-1), as could also be observed in the isothermal study (174.13 – 148.34 kJ mol-1 ), representing approximately a 30% difference in activation energy. These results confirm the poor stability of the prototype in the presence of lactose corroborating the results obtained in a compatibility study by thermal analysis and spectroscopy in the IR region (SALVIO-NETO; MATOS. May 2011).
CONCLUSION
The study of thermal behavior using thermal analytical techniques employed in this study allowed us to provide detailed information about a new chemical entity even when in its early stages of discovery from identification up to the development of a formulation. The differential scanning calorimetry allowed for the identification of LPSF/FZ4 through its band and melting enthalpy, while still providing quantitative data about its purity.
The comparison between the isothermal and non-isothermal methods showed good agreement for values of kinetic parameters (activation energy) demonstrating high thermal stability of the prototype. The decomposition kinetics occurred at a constant speed, order one (a single reaction that contributes to weight loss), i.e., there is only one significant reaction occurring and it is not affected by concurrent processes.
The results of the compatibility of certain binary blends were studied using TG/DTG, DSC, IR and decomposition kinetics. The results showed the usefulness of thermal analysis as a fast and convenient method in carrying out a screening of excipient candidates during preformulation studies, because it allowed the demonstration of interactions or drug-excipient incompatibility. In these studies, the possible interaction of LPSF/FZ4 with lactose was observed, confirmed by kinetic studies (isothermal and non-isothermal) which indicated a decrease of about 30% of the activation energy of degradation of pure prototype associated with lactose, showing a decrease in the stability of the drug.
Being thus, the present study contributed to the election of appropriate excipients for the development of a safe and steady formulation, exhibiting through the compatibility and the kinetic results that the association of LPSF/FZ4 with lactose is not recommended for the development of a solid pharmaceutical form.
So, the use of thermal analysis translates to an alternative of indubitable interest to the pharmaceutical field with the objective to quickly, with low cost, foresee the long term stability of a candidate for a pharmaceutical input and to accelerate its commercialization.
